Calorie restriction-mediated restoration of hypothalamic signal transducer and activator of transcription 3 (STAT3) phosphorylation is not effective for lowering the body weight set point in IRS-2 knockout obese mice

Abstract: Aim/hypothesis Lowering the body weight set point is a prerequisite for the maintenance of reduced body weight. In this context, obesity is known to be strongly linked to leptin resistance, and it remains to be clarified whether recovery from leptin resistance might lower the body weight set point to allow sustained body weight loss. Methods Obese IRS-2 knockout (IRS-2 -/-) mice were subjected to calorie restriction (CR) or b3-adrenergic receptor (AR) agonist treatment. The physiological effects of leptin, hyp… Show more

“…The cellular events in muscle of diabetic rats lead to the state of decreased glucose uptake and insulin resistance, which elicits further exaggeration of imbalanced glucose metabolism . On binding to IRS proteins, PI3K is activated, and then PI3K/Akt pathway is involved in insulin‐stimulated increase of glucose transport . Furthermore, on activation, GSK‐3β would also conduce to the consumption of glucose, which suggested the upregulation of GSK‐3β benefits in the recovery of patients with diabetes .…”

“…Traditional medicinal practices formed the basis of most of the early medicines followed by subsequent clinical, pharmacological, and chemical studies. 11 - 17 , 40 - 46 Plants have been well-documented for their medicinal uses for thousands of years. 47 They have evolved and adapted over millions of years to withstand bacteria, insects, fungi, and weather to produce unique, structurally diverse secondary metabolites.…”
