Gelidium floridanum W.R. Taylor tetraspores are units of dispersal and are responsible for substrate attachment. This study aimed to examine evidence of direct interaction between germ tube formation and Golgi activity during tetraspore germination of G. floridanum. After release, the tetraspores were incubated with brefeldin A (BFA) in concentrations of 4 and 8 μM over a 6 h period. The controls and treatments were analyzed with light, fluorescence (FM4-64 dye) and transmission electron microscopy. In the control samples, the Golgi bodies were responsible for germ tube formation. In contrast, BFA-treated samples were observed to inhibit spore adhesion and germ tube formation. These tetraspores also showed an increase in volume (≥30 μm width). BFA treatment also resulted in the disassembly of Golgi cisternae and the formation of vesiculated areas of the cytoplasm, blocking the secretion of protein and amorphous matrix polysaccharides. When stained with FM4-64, the control samples showed fluorescence in the apical region of the germ tube, but the treated samples showed an intense fluorescence throughout the cytoplasm. From these results, we can conclude that the germ tube is formed by the incorporation of vesicles derived from Golgi. Thus, vesicle secretion and Golgi organization are basic processes and essential in adhesion and tube formation. By blocking the secretion of protein and amorphous matrix polysaccharides, BFA treatment precluded tetraspore germination.

Kappaphycus and Eucheuma, known collectively as 'eucheumatoids', are two related genera of red seaweeds which currently lead the rankings for volume of global production of farmed macroalgae. Since 2009, the combined cultivated volume of these carrageenophytes overtook that of the brown seaweeds Laminaria (Saccharina) and Undaria for global production tonnages, according to statistics of the Food and Agriculture Organization of the United Nations (FAO). The Southeast Asian region, particularly Indonesia, the Philippines, Malaysia, Tanzania, and East Africa are the major producers of eucheumatoid biomass. Despite several success stories of red seaweed cultivation and the economic and socioeconomic value of their ecosystem services, there remain a number of salutary lessons to be learned from 'agronomic' practices applicable to their extensive cultivation. These case studies should be further developed, analysed, and adopted as best-practice recommendations for future socioeconomic prosperity, as well as both economic and environmental sustainability. In this review, we propose the use of the term 'phyconomy' (i.e. large-scale production of marine macroalgae for economic and industrial purposes) as an alternative to the term agronomy (i.e. terrestrial plant production).

Kappaphycus alvarezii (Doty) Doty ex P.C. Silva, a red macroalga, is a commercial source of carrageenan, a widely used polysaccharide compound important in the food and pharmaceutical industries, in nanotechnology, and in pharmacological applications. Carrageenan is found mainly in the cell wall and in the intercellular matrix. This is the first study to propose the characterization of carrageenans in vitro, using the auto-fluorescence properties of the alga treated with different polyamines: putrescine, spermidine, and spermine. This study suggests a four-phase cultivation sequence for seaweed farmers to enhance and assess the potential carrageenan yield of their crops. In phase 1, seedlings were treated with each of the polyamines. Explants were subsequently transferred through two additional culture phases before being planted on the sea farms in phase 4 and then harvested after 60 days for analysis. Images from transverse sections of 11 representative cultured K. alvarezii samples were obtained at 561 nm excitation wavelength for both the cell center and the cell wall of each sample. Spectral data were also analyzed using the spectral phasor algorithm of SimFCS developed at the Laboratory for Fluorescence Dynamics (www.lfd.uci.edu). We report on the identification of several spectral fluorescence emission fingerprints from different auto-fluorescence compounds spatially mapped using this technique. These fingerprints have the potential to improve strain selection of explants for enhanced carrageenan yield in seaweed farming operations as well as to enable wholesale pricing to correspond with crop quality.
