Calcium Ions Positively Modulate Follicle-Stimulating Hormone- and Exogenous Cyclic 3',5'-Adenosine Monophosphate-Driven Transcription of the P450scc Gene in Porcine Granulosa Cells

Abstract: Given the evident modulation of FSH-induced steroidogenesis by Ca 2ϩ in granulosa cells, we here test the hypothesis that Ca 2ϩ controls expression of the enzymatically rate-limiting cytochrome P450 scc (CYP11A) gene. To test this postulate, we quantitated the ability of Ca 2ϩ to regulate: 1) transcriptional activity of a transiently transfected luciferase reporter gene driven by a 2.32-kb 5Ј-upstream fragment of the porcine P450 scc gene promoter region; and 2) accumulation of endogenous P450 scc transcripts … Show more

“…Because hCG and FSH induce Ca 2ϩ changes in granulosa cells [16][17][18], these gonadotropins (which are the normal tropic hormones for the ovary) could also be used as positive controls. However, the Ca 2ϩ response of granulosa cells to FSH stimulation was not as consistent as the response to DDE.…”

“…DDT at concentrations of 64 and 128 M inhibited ATP-induced Ca 2ϩ uptake in bovine oviductal cells [15]. Calcium has been implicated in the mechanism of action of FSH in stimulating progesterone formation in rat [16] and porcine [17] granulosa cells, and hCG is a stimulator of Ca 2ϩ uptake in human granulosa cells [18]. The inhibitory effects of DDT on Ca 2ϩ uptake in bovine oviductal cells [15] and the effects of its metabolite DDE on thinning of egg shells [14] led us to hypothesize that DDE inhibits Ca 2ϩ uptake in human granulosa cells.…”

“…However, it is also expected that cellular processes activated by purinergic stimulation, and consequent release of intracellular Ca 2ϩ might have important effects on the metabolism of both the cumulus cell and oocyte compartments. It is important to mention that steroidogenesis, a principal functional role of granulosa cells, is affected by changes in the levels of both intracellular Ca 2ϩ and Cl Ϫ ions [52,53]. Also, it has been shown that intracellular Ca 2ϩ has a central role in the regulation of important processes in the oocyte [54].…”

“…This increase in Ca 2ϩ results in the formation of inositol triphosphate, which triggers the PKC signal cascade [33]. An increase in the uptake of extracellular Ca 2ϩ potentiates the effects of FSH and 8-Br-cAMP on transcription of the P450scc gene and progesterone synthesis in primary cultures of porcine granulosa cells [34]. Therefore, it is reasonable to speculate that the low concentrations of Cd 2ϩ used in the present study may be sufficient to mimic the effects of Ca 2ϩ , resulting in stimulation of the steroidogenic pathway and synthesis of progesterone in the JC-410 cells.…”
