Calcium Homeostasis and Reactive Oxygen Species Production in Cells Transformed by Mitochondria from Individuals with Sporadic Alzheimer’s Disease

Abstract: Alzheimer's disease (AD) is associated with defects in mitochondrial function. Mitochondrial-based disturbances in calcium homeostasis, reactive oxygen species (ROS) generation, and amyloid metabolism have been implicated in the pathophysiology of sporadic AD. The cellular consequences of mitochondrial dysfunction, however, are not known. To examine these consequences, mitochondrially transformed cells (cybrids) were created from AD patients or disease-free controls. Mitochondria from platelets were fused to 0… Show more

“…Cytosolic Ca 2ϩ concentrations ([Ca 2ϩ ]i) in endothelial cells were measured using fluorescent dye fura 2, as described (12). Briefly, cells cultured in 35-mm dishes were made quiescent by incubating overnight in serum-free medium.…”

“…After 1 hr the KRH buffer containing excess DCF was removed from the cells and replaced with fresh KRH and were treated by paraquat and inhibitors as described in figure legends. Fluorescent intensity was measured at 485 nm excitation and 520 nm emission on a Packard Fusion plate reader (Sheehan et al, 1997;Wang and Joseph, 1999). The KRH buffer was used in this assay because of high fluorescent background when using other medium e.g.…”

“…33 All cybrids of these diseases show some functional abnormalities. [34][35][36][37] Cybrid analysis is a powerful tool in studying the functional consequences of specific mtDNA mutations. However, its role in studies of neurodegenerative disorders is unclear and it is difficult to explain why these cybrids with mtDNA of platelets taken from patients have disease-specific abnormalities even though these diseases are not caused by specific mtDNA mutations.…”
