Calcium Flux and Contractility in Guinea Pig Atria

Abstract: A~STRACT The calcium in guinea pig atria can be divided into three components by kinetic studies with Ca4S: (a) a rapidly exchangeable fraction with a half-time of 4.5 minutes; (b) a slowly exchangeable fraction with a half-time of 86 (or 168) minutes; and (c) an inexchangeable fraction. In Krebs-Henseleit solution containing 2.5 m~ calcium, the calcium content of the tissue at rest remains constant, the flux being about 0.02 #t~mol/cm2-second. An increase or a decrease in extracellular calcium concentration b… Show more

“…Their hearts were rapidly removed and placed in a vessel containing heparinized Krebs-Henseleit (Krebs) solution (Winegrad & Shanes, 1962) previously gassed with 95% 02, 5% CO2. The composition of the solution was: NaHCO3 27-2 mM, NaCl 1180 mM, KCl 4-8 mM, KH2PO4 10 mM, MgSO47H20 1-2 mM, CaCl2-2H20 2-5 mm and glucose 11-1 mm.…”

“…Total calcium of the solution of the ash and total calcium of the nitric acid were measured with a Beckman DU spectrophotometer with a photomultiplier and an oxygen-hydrogen flame (3). The intensity at 556 m/u was used as a measure of calcium.…”

“…Small amounts of calcium are necessary for maximum contraction and maximum adenosine-triphosphatase activity in muscle models (1,31) Associated with contraction of intact muscle fibers there is an accelerated exchange of calcium probably involving both membrane and intracellular calcium (2,3). Hill has shown on theoretical grounds that insufficient time exists between membrane depolarization and muscle activation for a substance to diffuse from the cell surface to the cell center (4).…”

“…However, the interpretation ofexperiments designed to measure Ca2+ fluxes by using 'Ca2+ has been equivocal. Experiments with heart tissue have shown beat-dependent 'Ca2+ uptake (9)(10)(11)(12)(13)(14) but of a magnitude far greater than found in cultures of beating neonatal cells (15). In all cases, a major problem has been that the cellular Ca2+ represents such a small fraction of the total Ca2+ in the tissue that it is almost impossible to detect.…”
