Calcium Dobesilate: Pharmacological Profile Related to Its Use in Diabetic Retinopathy

Abstract: SUMMARYCalcium dobesilate (Doxium®) is used in the treatment of diabetic retinopathy. Clinical studies show a slowdown of the progression of the disease after long‐term oral treatment. The main action of the drug is related to a reduction of microvascular permeability as measured by different parameters and methods (vitreous fluorophotometry, retinal haemorrhages, skin capillary resistance, blood albumin leakage, blood viscosity) leading to improved visual acuity. The pharmacological activity may be explained … Show more

“…It is widely recognized that phlebotonic activity extends beyond the mere elevation of venous wall tone. In the modern context, veno-active drugs should possess the ability to stimulate lymphatic drainage and enhance microcirculation [80], employing various mechanisms [81][82][83][84]. Endothelial dysfunction holds significance within the venous system as it may lead to a decrease in venodilation, resulting in an increase in venous tone and a decrease in venous compliance.…”

“…Calcium dobesilate ( 1 ) is an orally administered angioprotective agent for reducing blood, plasma, and aqueous humor hyperviscosity and microvascular hyperpermeability. Up to approximately 60% incidence in developed countries use 1 as the oral treatment for diabetic retinopathy, hemorrhoidal syndrome, and chronic venous insufficiency in the lower limbs (pain, cramps, paresthesia, edema, and stasis dermatosis). After being prescribed worldwide for many years, possible conferring benefits have been discovered including potential anticancer and antiviral effects. Additionally, recent investigations have shown that 1 is an effective therapy for COVID-19 patients because its capacity to inhibit several signaling pathways can implicate COVID-19 pathophysiology. , …”
