Calcium-Dependent NOX5 Nicotinamide Adenine Dinucleotide Phosphate Oxidase Contributes to Vascular Oxidative Stress in Human Coronary Artery Disease

Abstract: Objectives To examine the expression and activity of the calcium dependent NADPH oxidase in human atherosclerotic coronary arteries. Background The Nox based NADPH oxidases are major sources of reactive oxygen species (ROS) in human vessels. Several Nox homologs have been identified but their relative contribution to vascular ROS production in coronary artery disease (CAD) is unclear. Nox5 is a unique homolog in that it is calcium dependent and thus could be activated by vasoconstrictor hormones. Its presenc… Show more

“…NOX5 has been reported to be induced by thrombin (34), Ang II (300), and platelet-derived growth factor (197). An increased expression level of NOX5 has been implicated in the coronary artery disease patients, which correlates with the oxidative damage observed in atherosclerosis (157). NOX5 has also been reported to be essential for PDGF-stimulated proliferation of vascular smooth muscle cells and proliferation of endothelial cells and angiogenesis (34,197).…”

“…ROS production is increased in VSMCs from resistance arteries of hypertensive patients, and this is associated with up-regulation of vascular Nox (75,112,142,207,213,233). The importance of Nox in human cardiovascular disease is supported by studies showing that polymorphisms in Nox subunits are associated with increased atherosclerosis and hypertension.…”

“…These include Nox1, Nox2 (formerly gp91phox), Nox3, Nox4, Nox5, Duox1 and Duox2. [73][74][75] All Noxes are transmembrane proteins that have conserved structural properties and that transport electrons across biological membranes to reduce O 2 to O 2 À . Nox1, Nox2, Nox4 and Nox5 have been identified in vascular tissue.…”
