Calcium Coordination Solids for pH‐Triggered Release of Olsalazine

Abstract: Calcium coordination solids were synthesized and evaluated for delivery of olsalazine (H4olz), an anti‐inflammatory compound used for treatment of ulcerative colitis. The materials include one‐dimensional Ca(H2olz)⋅4 H2O chains, two‐dimensional Ca(H2olz)⋅2 H2O sheets, and a three‐dimensional metal‐organic framework Ca(H2olz)⋅2DMF (DMF=N,N‐dimethylformamide). The framework undergoes structural changes in response to solvent, forming a dense Ca(H2olz) phase when exposed to aqueous HCl. The compounds Ca(H2olz)⋅x … Show more

“…Olsalazine (H 2 OLZ), a prodrug of the anti-inflammatory 5-aminosalicylic acid (5-ASA), is prescribed as the first line of treatment for patients with idiopathic inflammatory bowel diseases such as Crohn's disease and ulcerative colitis. [122] To solve the problem regarding sustained release of olsalazine in human body, Long and co-workers [123] (3). After being pressed into pellets and exposed to simulated gastrointestinal fluids, all three materials exhibited a delayed release of olsalazine relative to Na 2 (H 2 OLZ) which released more than 90% olsalazine.…”

“…Calcium, the fifth most abundant element in the earth’s crust, fills many important roles in natural and synthetic chemistry . Ca 2+ is the main cation in bone, shell, and microstructured biomaterials with remarkable mechanical properties. It is often employed as an enzymatic cofactor, including within the photosynthetic oxygen-evolving complex (OEC), and is considered “the best communicator” in biological signaling. Calcium is a key component in cements and many industrial chemical processes, and molecular calcium complexes can effect alkene and imine hydrogenations, polymerizations, and even the nucleophilic alkylation of benzene. Partly because it is such a “green” metal compared to the often rare or toxic stalwarts of transition-metal chemistry, interest in calcium catalysts and metal–organic frameworks is increasing. ,, …”
