Calcium and Magnesium Binding to Human Centrin 3 and Interaction with Target Peptides

Abstract: There are four isoforms of centrin in mammals, with variable sequence, tissue expression, and functional properties. We have recently characterized a number of structural, ion, and target binding properties of human centrin isoform HsCen2. This paper reports a similar characterization of HsCen3, overexpressed in Escherichia coli and purified by phase-reversed chromatography. Equilibrium and dynamic binding studies revealed that HsCen3 has one mixed Ca(2+)/Mg(2+) binding site of high affinity (K(d) = 3 and 10 m… Show more

“…In human Centrin2 and Scherffelia dubia centrin, the N-terminal extension has been shown to be required for Ca 2+ -induced centrin self-assembly (Tourbez et al 2004;Wiech et al 1996;Yang et al 2006). However, in yeast centrin CDC31, the N-terminal extension is not essential for CDC31 function in vivo (Li et al 2006), though Ca 2+ - Cox et al (2005) The experimental methods used to determine the structural properties are indicated in brackets induced polymerization is observed in vitro at high CDC31 concentrations (Wiech et al 1996).…”

“…HsCen-2 Metal Binding and EF-hands-The affinities of Ca 2ϩ -binding sites within HsCen-2 and other centrins are lower than for calmodulin (43,(47)(48)(49) Table 2 herein. 27-fold higher affinity for the XPC peptide used herein compared with Ca 2ϩ -free centrin-2 (11).…”

“…ME bond with EoCen at the ratio of 1 : 1 under the experimental conditions. Different from Human centrin 3 (HsCen3), both HsCen2 and EoCen formed 1 : 1 centrin-peptide complex [10] . Similar conclusion has been reported on Chlamydomonas reinhardtii caltractin (CRC) that it is capable of binding simultaneously two amphiphilic peptides derived from yeast Kar1p protein [11] .…”
