Calcineurin is essential for DNA synthesis in Swiss 3T3 fibroblasts

Abstract: DNA synthesis was measured 16 h after stimulation of Swiss 3T3 fibroblasts in the resting phase with various growth factors (platelet-derived growth factor, fibroblast growth factor, lysophosphatidic acid and thrombin). When extracellular Ca2+ was chelated by EGTA, or when the influx of Ca2+ from outside to inside the cell was blocked by cobalt, DNA synthesis was completely inhibited. As there was no effect whatsoever on DNA synthesis when Ca2+ was chelated, or when the influx of Ca2+ was blocked up to the fir… Show more

“…Because protein synthesis is required for cells to exit from G 0 and enter the cell cycle, an effect of the immunosupressants could also block mitogenesis. In fact, both CsA and FK506, but not a Ca 2ϩ /calmodulin-dependent protein kinase II inhibitor, inhibited DNA synthesis in fibroblasts (46). At present, however, there is no obvious mechanism by which CN might regulate the translation machinery.…”

“…In mammals, calcineurin mediates T cell and B cell proliferation (15,24,25,36) and has been implicated in urotensin II-mediated proliferation of airway SMC (7), angiotensin II-induced cardiac fibroblast hyperplasia (12), and pituitary cell proliferation (11). Indeed, calcineurin has been shown to be essential for DNA synthesis in Swiss 3T3 fibroblasts (47), where inhibitors of calcineurin block the fibroblast growth factor-mediated expression of cyclins A and E (48).…”

“…Both c-Myb and calcineurin influence Ca 2ϩ -mediated signals of cell cycle (1,22,47,48 influx and a c-Myb-dependent reduction in Ca 2ϩ efflux mediated via repression of PMCA1 (1,22,23,40,41). Indeed, ϳ60% of the c-Myb effect on VSMC proliferation is exerted via PMCA1 repression (22).…”

“…1 In several cell types, DNA synthesis is completely inhibited when extracellular Ca 2+ was chelated by EGTA, or when the influx of extracellular Ca 2+ was blocked by cobalt. 2,3 Similarly, depletion of extracellular Ca 2+ during the first 8 h after growth factor stimulation resulted in an inhibition of DNA synthesis in human fibroblasts. 4 In addition, depletion of intracellular Ca 2+ stores with pharmacological agents, such as the sarco-endoplasmic reticulum Ca 2+ ATPase (SERCA) inhibitor thapsigargin, resulted in significant abrogation or delay of progression to S phase.…”
