Calcineurin-GATA4 Pathway Is Involved in β-Adrenergic Agonist-responsive Endothelin-1 Transcription in Cardiac Myocytes

Abstract: Increases in the expression of endothelin-1 (ET-1) in cardiac myocytes play a critical role in the development of heart failure in vivo. Whereas norepinephrine (NE) is a potent inducer of ET-1 expression in cardiac myocytes, the signaling pathways that link NE to inducible cardiac ET-1 expression are unknown. Adrenergic stimulation results in an increase in intracellular calcium levels, which in turn activates calcineurin. Here, we have shown that stimulation with NE markedly increased the expression of the ET… Show more

“…p300 acetylates not only histone, but also hypertrophy-responsive transcription factors by forming a huge complex. 16,26,27,35 The present study demonstrated that spironolactone almost completely inhibited PEinduced cardiomyocyte hypertrophy. However, our findings do not rule out possible roles of pathways other than MRdependent ones in the hypertrophic response.…”

“…In endothelial cells, ET-1 production is regulated at the level of transcription, mRNA stability, and maturation of the ET precursor protein, preproendothelin. Whereas extracellular ET-1 reduces the cellular content of ET-1 mRNA in endothelial cells [74], Ang II, oxidized LDL, insulin, isoproterenol, thrombin, TGFβ, TNFα, verotoxin, and VEGF increase ET-1 mRNA abundance [75][76][77]. The best characterized intracrine signalling system is that of peptidergic agonist, Ang II.…”

“…These factors interact with GATA4 and enhance gene transcription. [48][49][50] Friend of GATA (FOG)-2 is a transcriptional co-factor, and its interaction with GATA4 competes with the interaction of GATA4 with p300 and results in the repression of target genes in cardiac myocytes 51) . GATA6 is a member of the GATA family and is expressed in cardiomyocytes.…”
