Calcimimetic R-568 or calcitriol: equally beneficial on progression of renal damage in subtotally nephrectomized rats

Abstract: Patients with renal insufficiency develop secondary hyperparathyroidism. Monotherapy with active vitamin D or calcimimetics ameliorates secondary hyperparathyroidism. We compared kidney damage in subtotally nephrectomized (SNX) rats treated with active vitamin D (calcitriol) or the calcimimetic R-568. Male Sprague-Dawley SNX and sham-operated (sham-op) rats were randomized into the following treatment groups: SNX ϩ R-568, SNX ϩ calcitriol, SNX ϩ vehicle, sham-op ϩ R-568, sham-op ϩ calcitriol, and sham-op ϩ veh… Show more

“…The experiment was terminated 8 or 12 wk after the surgery under general anesthesia (100 mg/kg ketamine hydrochloride, Ketamin, 10%; Essex, Tierarznei, Germany) and 3.0 mg/kg xylazine (Xylazin, 2%; Ceva, Tiergesundheit, Germany). The abdominal aorta was catheterized, blood samples were taken, and retrograde perfusion fixation was performed using 3% glutaraldehyde solution at body temperature for morphological investigations or ice-cold NaCl for immunohistochemistry and Western blotting, respectively (21). The perfusion pressure was kept constant at 120 mmHg.…”

“…The number of glomeruli per kidney and mean glomerular volume were determined using the point counting method as previously described (21). Capillaries and the number and volume of glomerular cells were analyzed in semithin sections at ϫ1,000 magnification using the point-counting method as previously described (21).…”

“…) and calcitriol were equally beneficial in slowing the progression of renal damage in subtotally nephrectomized rats, which exhibited marked secondary hyperparathyroidism when fed a normal phosphate diet (29). The study used 12-wk-old Sprague-Dawley rats and weight-controlled nephrectomy.…”

“…Besides, any reduction in UACR must also be interpreted in the context of its biological variation in patients with diabetes, which is estimated at 61%. 2 Second, the mean concentration of vitamin D in the patients included (40 nmol/L) is far below the current recommendations 3,4 and refl ects a severe defi ciency. Subgroup analysis according to the 25-hydroxyvitamin D concentration would be of interest.…”
