Calbistrins, novel antifungal agents produced by Penicillium restrictum. I. Production, taxonomy of the producing organism and biological activity.

Abstract: A novel antibiotic complex, named the calbistrins, has been discovered in the culture broth of a soil fungus. The producing organism, designated AB1875C-28, was identified as a strain of Penicillium restrictum. Calbistrin A, the most potent of the 4-membered complex, has MICsof 0.78 /zg/ml against Candida albicans. Only poor activity is observed against non-candida yeasts, filamentous fungi and bacteria.

“…As far as we are aware this is the first report of P. oxalicum producing an antifungal substance containing least two active compounds. Fungi of the genus Penicillium are important producers of antibacterial compounds, but only a few are reported to produce antifungal substances (Kumagai et al 1990;Matsukuma et al 1992;Jackson et al 1993;Okada et al 1998;Nose et al 2000;Sasaki et al 2000;Kaiserer et al 2003;He et al 2004;Nicoletti et al 2004). HPLC separation results showed that strain PY-1 produced at least two active compounds which may significantly inhibit mycelial growth of many pathogenic fungi and suppress infection by S. sclerotiorum when aerially applied.…”

“…To our knowledge, the nature and chemical composition of antifungal compounds produced by EU0013 has not been studied. The genus Penicillium produces both antibacterial (Kumagai et al 1990;Matsukuma et al 1992;Jackson et al 1993), and antifungal compounds (Yang et al 2008). In environmentally controlled experiments using sterile potting medium, EU0013 significantly suppressed Fusarium wilt in tomato and cabbage.…”

“…Compound 3 also inhibited aromatase with an IC 50 value of 12.3 lM (Stresser et al, 2000), a useful therapeutic target for the treatment of breast cancer (Brueggemeier et al, 2005). Previously, calbistrins A-D were found to exhibit potent antifungal activity, particularly calbistrin A which displayed MIC values of 0.78 lg/mL (Jackson et al, 1993).…”
