The roles of subcellular distribution on silver (Ag) accumulation and elimination in the green mussel Perna viridis collected from 2 hydrographical zones (harbour vs. estuarine) in Hong Kong during 3 different seasons were investigated. Ag concentration and biokinetics varied widely between the 2 zones, but were less variable among the different seasons (pre-wet, wet and dry). The harbour population had a 2-to 6-fold higher Ag tissue concentration than the estuarine population. The insoluble fraction was the predominant pool for Ag subcellular distribution in the green mussels. In both the pre-wet and wet seasons, a lower Ag tissue concentration was associated with the insoluble fraction in the estuarine population as compared to the harbour population. Ag uptake from the aqueous phase was faster in the estuarine population during all seasons, but no significant relationship between Ag uptake and salinity was found in this study. Dietary uptake of Ag was similar in both populations, although there was a slight difference in assimilation from diatoms and natural seston. Efflux of Ag was 2 times faster in the estuarine population during the wet and dry seasons, which may partially explain the lower Ag tissue burden in the mussels. These results demonstrated a close correlation between Ag kinetics and its subcellular distribution. The percentage of Ag in the heatsensitive proteins correlated positively to the influx of dissolved Ag and Ag efflux, but negatively to the dietary assimilation. On the other hand, a higher percentage of Ag in the insoluble fraction was related to a higher dissolved uptake rate constant and a lower assimilation. Thus, the role of the individual subcellular fraction in the uptake was exposure pathway-specific. We examined the dynamics of subcellular distribution of metals (Cd, Ag, and Zn) in the marine green mussel Perna viridis by partitioning the metals into the insoluble fraction (IF), heat-sensitive proteins (HSP), and metallothionein-like proteins (MTLP) during metal uptake and elimination. Variations in metal uptake and elimination then were correlated with the subcellular distributions of these metals. The IF and HSP were the first ligands to bind with the metals during the dissolved exposure, and more metals were found in the HSP when the metal influx rate was higher. However, to minimize toxicity, metals were redistributed from HSP to MTLP afterwards. The subcellular distribution of metals was dependent of the exposure route in the mussels. During dietary metal exposure, the metals attained equilibrium before they were assimilated and the metal assimilation efficiency was independent of the metal partitioning in different subcellular fractions. During the efflux, metals in the soluble fraction mediated depuration, whereas metals in the insoluble fraction acted as a final storage pool. Redistribution also may occur between the metal-sensitive and inactive pools without significant depuration as a secondary protective mechanism. We further demonstrated that the higher efflux rate of Ag and Cd was related to a higher partitioning in the MTLP and a lower partitioning in the IF. Our study shows that subcellular pools other than MTLP were involved in immediate metal handling in the bivalves. The wide dynamics of subcellular metal distribution suggests that the relevance of individual subcellular fractions is dependent on the exposure pathway.

A very large number of experimental studies in aquatic ecotoxicology have been devoted to the assessment of metal uptake in different classes of living organisms. Most of them are based on analysis of single toxicants, despite the fact that under field conditions the biota is generally exposed to multicontaminant pollution. In addition to producing a direct response in biota, a toxic metal can also act by interfering with other metal ions or elements. Thus, the concurrent presence of two or more metallic contaminants in the organism often yields metal uptake that deviates from those currently observed for individual metals. If the effect is an enhancement of bioaccumulation, it is said to be synergistic. If the effect is a decrease of metal incorporation, it is said to be antagonistic.Most of the knowledge about metal-metal interactions has been acquired in mammalian species. In such species, the major source of metals is the diet and metal uptake depends on the physicochemical forms of storage of metals in food organisms (Amiard-Triquet et al., 1993;Dallinger, 1993). In these conditions, interactions occur inside the organism and changes may be expected at the level of toxicant uptake, distribution, storage and excretion. Biochemical processes such as competitive affinities for intracellular ligands or co-accumulation as mineral granules are probably predominant. In the case of aquatic organisms, interactions can occur both inside organisms (as in mammals) as well as outside through changes of metal speciation, distribution between soluble and insoluble phases and finally bioavailability. Great differences have been detected in metal body burdens when different species (or even different populations within the same species) were compared (Rainbow, 1993).
