Ca2+ entry through mechanotransduction channels localizes BAIAP2L2 to stereocilia tips

Abstract: BAIAP2L2, a membrane-binding protein required for maintenance of mechanotransduction in hair cells, is selectively retained at tips of transducing stereocilia. BAIAP2L2 trafficked to stereocilia tips in the absence of EPS8, but EPS8 increased the efficiency of localization. A tripartite complex of BAIAP2L2, EPS8, and MYO15A formed efficiently in vitro, and these three proteins robustly targeted to filopodia tips when co-expressed in cultured cells. Mice lacking functional transduction channels no longer concen… Show more

“…BAIAP2L2 (also known as pinkbar) localizes to the tips of short stereocilia in cochlear hair cells, and both conditional and full knockout mouse mutants show progressive stereocilia degeneration and hearing loss (Carlton et al, 2021; Yan et al, 2021). BAIAP2L2 is transported to the tips in a complex with EPS8 and MYO15A (long isoform) and appears after the stereocilia have already formed (Carlton et al, 2021; Halford et al, 2022). Subsequently, MET channel activity is required to maintain BAIAP2L2 at the tips, possibly through Ca 2+ influx and/or binding to the channel subunit CIB2 (Halford et al, 2022; Yan et al, 2021).…”

Notably, BAIAP2L2 has a unique almost-flat membrane-binding surface, which may better suit it for the wide morphology of stereocilia.…”

“…Several proteins concentrate at tips of row 2 stereocilia in developing hair bundles, including EPS8L2, ESPNL, BAIAP2L2, CAPZB, TWF2, DSTN and CFL1, and the locations of most of these are regulated by mechanotransduction 33–38 . We examined the location of two of these, EPS8L2 and CAPZB, in Ush1g bw hair cells.…”
