C-terminal binding proteins: central players in development and disease

Abstract: C-terminal binding proteins (CtBPs) were initially identified as binding partners for the E1A-transforming proteins. Although the invertebrate genome encodes one CtBP protein, two CtBPs (CtBP1 and CtBP2) are encoded by the vertebrate genome and perform both unique and duplicative functions. CtBP1 and CtBP2 are closely related and act as transcriptional corepressors when activated by nicotinamide adenine dinucleotide binding to their dehydrogenase domains. CtBPs exert transcriptional repression primarily via re… Show more

“…The CTBP binding site in FOXP2 was previously suggested to be a PLNLV motif at residues 422–426, due to the similarity of this motif to the consensus CTBP-binding motif PXDLS (Fig. 4a) [24, 36]. The PLNLV motif is conserved in FOXP1 but not FOXP4, consistent with the lack of interaction between CTBP1 and FOXP4 [24] (Fig.…”

“…Relatively, little is known about the interaction partners of FOXP2; however, proteins of the CTBP family have been identified as candidate FOXP2 interactors in multiple independent yeast two-hybrid screens [24, 3234]. The CTBP family consists of two proteins, CTBP1 and CTBP2, which function as co-repressors for multiple transcription factors [35, 36]. The interaction between FOXP2 and CTBP1 has previously been validated by us using the BRET assay, and by others via co-immunoprecipitation, but the interaction with CTBP2 has not yet been confirmed [19, 24].…”

“…The most common scaffolding protein in the interaction network is TRIM28 (Fig. 5A), followed by TRIM27 (8 interactions), CTBP1 (four interactions), and CTBP2 (three interactions), all of which have been implicated in recruitment of histone modification complexes (Bloor et al 2005;Stankiewicz et al 2014). These findings support a widespread role of C2H2-ZF proteins in chromatin structure and organization.…”
