C/EBP transcription factors regulate SREBP1c gene expression during adipogenesis

Abstract: The transcription factor SREBP1c (sterol-regulatory-element-binding protein 1c) is highly expressed in adipose tissue and plays a central role in several aspects of adipocyte development including the induction of PPARγ (peroxisome-proliferator-activated receptor γ), the generation of an endogenous PPARγ ligand and the expression of several genes critical for lipid biosynthesis. Despite its significance, the regulation of SREBP1c expression during adipogenesis is not well characterized. We have noted that in s… Show more

“…Motif analysis at sites with stronger C/EBPα binding in the Trib1_LSKO mice revealed robust enrichment for the canonical C/EBP motif (Figure 7B), and gene-set enrichment analysis of these peaks showed major enrichment of genes involved in lipid metabolism as one of the top findings (Supplemental Figure 6A). Moreover, differ- thesis, such as ChREBP, LXR, and SREBP1, the latter of which C/EBPα is reported to regulate the transcription of in adipose (8). C/EBPα protein abundance in liver is regulated by TRIB1, and thus the TRIB1-C/EBPα axis may be an underappreciated moderator of human hepatic lipogenesis.…”

“…Among the targets of C/EBP and C/EBPd are the promoters of the genes encoding the key adipogenic transcription factors C/EBP and PPARg and the regulator of lipogenic genes SREBP1 (for sterol-regulatory-element-binding protein 1) (Payne et al, 2010;Rosen and MacDougald, 2006;White and Stephens, 2010). PPARg activates the promoter of the gene encoding C/EBP and vice versa, creating a positive-feedback loop.…”
