c-Abl regulates Mcl-1 gene expression in chronic lymphocytic leukemia cells

Abstract: IntroductionChronic lymphocytic leukemia (CLL) is a malignant disease characterized by the gradual accumulation of mature, nonproliferative B cells expressing a distinctive group of surface markers, including CD19, CD23, and CD5. 1,2 It is a heterogeneous disease that can follow an indolent or aggressive course, and understanding the nature of this heterogeneity is an area of intense research interest. 2 CLL cells are resistant to apoptosis, and this is thought to be regulated by survival signals from the micr… Show more

“…45 Moreover, inhibition or downregulation of Abl has been reported to reduce the expression of the anti-apoptotic protein Mcl-1 and induce leukemic cell apoptosis. [45][46] Although we did not observe a direct cytotoxic effect of VX-680 against resting CLL cells, it remains possible that this compound could enhance the sensitivity of these cells to other cytotoxic agents by down-regulating Mcl-1.…”

“…NF-kB, known to positively regulate Mcl-1, 28 is constitutively activated in CLL cells. 29 We examined the effects of 4HPR on NF-kB activity by western blot, because the antibody was not well suitable for flow cytometry.…”
