C-11 Radiochemistry in Cancer Imaging Applications

Abstract: Carbon-11 (C-11) radiotracers are widely used for the early diagnosis of cancer, monitoring therapeutic response to cancer treatment, and pharmacokinetic investigations of anticancer drugs. PET imaging permits non-invasive monitoring of metabolic processes and molecular targets, while carbon-11 radiotracers allow a "hot-for cold" substitution of biologically active molecules. Advances in organic synthetic chemistry and radiochemistry as well as improved automated techniques for radiosynthesis have encouraged i… Show more

“…For short-lived radionuclides such as C-11 and F-18, specific radioactivities are also much improved, and rapid radiolabeling is central to success. Comprehensive reviews are available that detail these challenges and provide practical solutions [6972]. The synthetic methods and special practical issues associated with incorporation of radioiodine [7376] and radiometals [7780] have also been addressed.…”

“…Finally, the organic nitrile function thus introduced can be converted to a variety of chemical modifications, allowing the preparation of a wide array of carbon‐isotope labeled compounds . While 14 C‐labeled compounds are an essential tool to get market access for crop protection products, and 14 C‐labeled active pharmaceutical ingredients are still the main radioactive markers for several pharmaceutical studies including human metabolism studies, carbon‐11 labeled compounds are widely used radiotracers for drug development …”

“…One major 11 C precursor used in the synthesis is [ 11 C]‐CO 2 (the other being [ 11 C]‐CH 4 ) . To date, the most frequently used method for the introduction of 11 C into organic molecules is the methylation . Methylations with this isotope are limited to the labeling of molecules at functional groups at peripheral atomic positions.…”
