Fusarium head blight (FHB) is a devastating disease of wheat. Worldwide, Fusarium graminearum is the most dominant FHB-causing species. Its most common toxin, deoxynivalenol (DON), impairs food and feed safety and has an enormous economic impact. Agronomic factors such as crop rotation, soil management and host genotype strongly influence the occurrence of F. graminearum. Infected plant debris from previous crops, on which perithecia and ascospores develop, represent the main source for FHB, and hence, improved cropping systems aim to reduce this inoculum to decrease the infection risk. The best measure to evaluate the disease pressure is spore traps that detect deposited airborne ascospores. Commercial spore traps are expensive and require power sources, thus, they are not suitable for investigations in field experiments with different treatments. In consequence, we developed spore traps containing a Petri dish with Fusarium-selective agar, protected by aluminum dishes and attached on a wooden board. We compared the data of our low-cost trap with those of a commercial high-throughput jet sampler and obtained equivalent results. In field experiments to compare cropping systems, we observed a high correlation between the DON content in wheat grains and the number of colonies from deposited spores. Our spore trap proved to be a highly valuable tool to not only study FHB epidemiology but also to identify innovative cropping systems with a lower risk for FHB and DON contamination.

Feeding the world's ever‐increasing population requires continuous development of high‐yielding and disease‐resistant cultivars of food crops such as wheat (Triticum aestivum L.). Speed breeding, which utilizes longer photoperiod times and higher temperatures, is a technique that accelerates plant development and is rapidly being adopted by wheat breeders across the globe to fast‐track cultivar development. Plant diseases are a major threat to crop production, and breeding for disease resistance is a major goal of crop breeders. Fusarium head blight (FHB), caused by Fusarium graminearum, is a major disease of small grain cereals, affecting their yield and quality. The aim of present work was to assess if speed breeding conditions can be used to accelerate reliable assessment of FHB severity and mycotoxin deoxynivalenol (DON) accumulation in wheat varieties. We screened a set of six spring wheat genotypes with different levels of genetic resistance (two moderately susceptible, two highly susceptible, one moderately resistant, and one resistant) for their response to FHB at 14 days after inoculation (dai) and 21 dai and DON accumulation under normal versus speed breeding conditions. FHB severity and DON accumulation were found to be highly correlated at all time points under normal and speed breeding conditions. Robust differentiation between resistant and susceptible genotypes could be achieved at 14 dai rather than the normal period of 21 dai, saving at least a week in phenotyping. Combined with the accelerated growth, flowering, and maturity under these conditions, efficient FHB screening and DON evaluation under speed breeding conditions will fast‐track development of resistant wheat varieties.

Different diagnostic approaches for the characterization of the fungal community and Fusarium species complex composition of Italian durum wheat grain and correlation with secondary metabolite accumulation

BACKGROUND The evolution of the fungal communities associated with durum wheat was assessed using different diagnostic approaches. Durum wheat grain samples were collected in three different Italian cultivation macro‐areas (north, center and south). Fungal isolation was realized by potato dextrose agar (PDA) and by deep‐freezing blotter (DFB). Identification of Fusarium isolates obtained from PDA was achieved by partial tef1α sequencing (PDA + tef1α), while those obtained from DFB were identified from their morphological characteristics (DFB + mc). The fungal biomass of eight Fusarium species was quantified in grains by quantitative polymerase chain reaction (qPCR). Fungal secondary metabolites were analyzed in grains by liquid chromatography–tandem mass spectrometry (LC–MS/MS). Correlations between Fusarium detection techniques (PDA + tef1α; DFB + mc and qPCR) and mycotoxins in grains were assessed. RESULTS Alternaria and Fusarium showed the highest incidence among the fungal genera developed from grains. Within the Fusarium community, PDA + tef1α highlighted that F. avenaceum and F. graminearum were the most represented members, while, DFB + mc detected a high presence of F. proliferatum. Alternaria and Fusarium mycotoxins, principally enniatins, were particularly present in the grain harvested in central Italy. Deoxynivalenol was mainly detected in northern‐central Italy. CONCLUSIONS The adoption of the different diagnostic techniques of Fusarium detection highlighted that, for some species, qPCR was the best method of predicting their mycotoxin contamination in grains. © 2023 The Authors.
