Building digital twins of the human immune system: toward a roadmap

Abstract: Digital twins, customized simulation models pioneered in industry, are beginning to be deployed in medicine and healthcare, with some major successes, for instance in cardiovascular diagnostics and in insulin pump control. Personalized computational models are also assisting in applications ranging from drug development to treatment optimization. More advanced medical digital twins will be essential to making precision medicine a reality. Because the immune system plays an important role in such a wide range o… Show more

“…It may revolutionise the management of obesity and its comorbidities, and provide a pillar for healthy ageing. Moreover, Laybenbacher et al 63 outlined a roadmap for building an immune DT to capture key features of the immune system, which was to structure in four stages: specification, model constructions, personalisation and continued improvement. It would help to change the nature of biomedical research and accelerate the clinical translation of basic research.…”

“…One of the primary benefits of in silico clinical trials is the capacity to run experiments in a virtual environment without the need for expensive and timeconsuming real-world experiments. One technology being heavily discussed in the clinical trial space is the idea of a digital twin -an artificial physiological model of a patient that responds to various clinical trial experiments [65,66].…”

“…However, digital twins are a relatively novel idea in the clinical space. While there has been much work in the form of position papers promoting the general concept of digital twins [68,69,70,71,66], only a few have described their methodology in detail.…”

“…There are increasing interests and efforts in the space of digital twinning of patients ( 10 , 1 ). And we believe that amongst the most natural, feasible and on-the-near-immediate-horizon approaches to digital twins are so-called theranostic digital twins (TDTs), which we describe next.…”
