Broxaterol (Z.1170), a New Oral <i>β</i>2-Agonist Compared with Salbutamol

Abstract: The bronchodilating activity and tolerability of a single 0.5-mg oral dose of broxaterol (Z.1170) were evaluated in 18 patients with reversible bronchial obstruction. Salbutamol 4.0 mg and placebo were used as controls. The study design was double-blind within patients. The forced expiratory volume in 1 s (FEV1), pulse rate, and blood pressure were measured immediately before and 0.5, 1, 2, 3,4, 5, and 6 h after each treatment. At the same time clinical controls were made to detect the possible pres… Show more

“…In this respect, a comparison was made between salbutamol and broxaterol, a new Effects o f fT-Agonists d u rin g Exercise in C h ro n ic A irflow L im itatio n selective P^-sympathomimetic agent which in vitro has shown a higher affinity than that of salbutamol for (32-adrenoceptors and a very low intrinsic activity on pr adrenoceptors [18],…”

“…An other study compared only one dose of each drug, and showed that an 8 times higher dose of salbutamol gave the same bronchodilation as broxaterol [7], How ever, the present study is the only one which has been performed with cumula tively increasing doses of the drugs. This study showed a considerable increase of heart rate with oral treatment of both broxaterol and salbutamol.…”

“…This study showed a considerable increase of heart rate with oral treatment of both broxaterol and salbutamol. Earlier studies have also shown effects on heart rate, with higher doses of broxaterol, mainly 0.5 mg, similarto 4 mg salbutamol, also indicating that the clinical selectivity for p2-adrenoreceptors are the same for the two drugs [6][7][8],…”

“…Recently, in patients with reversible airway obstruction Perruchoud et al [13] reported no significant difference between the effects of 0.5 mg broxaterol and those of 4 mg salbutamol, both orally adminis tered, and they underlined the good toler ance of the two drugs. Perruchoud's re sults confirm a previous report of Rampulla et al [1] and Blasi and Pezza [2].…”
