1 The effects of a single oral dose of the dopaminergic agonists lisuride (0.1 mg), terguride (0.25 mg) and bromocriptine (1.25 mg) on IOP were studied in eight normal volunteers using the non-contact tonometer. 2 Considering all post-dose measurements, compared with placebo, bromocriptine and lisuride but not terguride reduced IOP significantly in both eyes. 3 There was no significant difference between the ocular hypotensive effect of bromocriptine and lisuride. 4 Terguride reduced IOP significantly in the left eye at the 3 h time point after drug administration. 5 The result of this study confirms the reported ocular hypotensive effect of bromocriptine and showed that lisuride is as effective as bromocriptine in reducing IOP. 6 To evaluate the clinical importance of these drugs as ocular hypotensive agents other studies are needed using eye drops.

1. The ocular hypotensive effect of a single oral dose of 25 micrograms pergolide, a dopamine 2‐receptor agonist, was studied in 12 normal human volunteers, using a non‐invasive method. 2. An oral dose of timolol 20 mg was used as a positive control. 3. The effects of both drugs on pupil diameter were also studied, using a photographic method. 4. Considering the first 6 h post‐dosing measurements, using multiple linear regression analysis comparing drug with placebo and including the baseline values as continuous independent variables, both pergolide and timolol had a significant ocular hypotensive effect in both eyes (P less than 0.0001) with no significant effects on pupil diameter. 5. Further topical application studies in normal and glaucomatous eyes are needed to evaluate the usefulness of pergolide in the treatment of glaucoma.

SUMMARYThe interaction between bromocriptine and metoclopramide on intraocular pressure (IOP), pupil diameter, and other parameters were studied in healthy volunteers. Pupil diameter was not affected by both treatments and only bromocriptine lowered IOP. Prior treatment with metoclopramide blocked the IOP-lowering effect of bromocriptine. It is concluded that bromocriptine lowers IOP by stimulation of dopamine-2 receptors.Oral bromocriptine (1-25 mg single dose) lowers intraocular pressure (IOP) in healthy volunteers without affecting blood pressure or pupil diameter.' Materials and methodsEight healthy male volunteers participated in this study. None wore contact lenses. They attended on four occasions, at least seven days apart, at the same time of the day (10-30-11-00 am) to receive either metoclopromide (10 mg) or matched placebo intravenously followed (15 minutes later-time zero) by either bromocriptine (1-25 mg) or a matched placebo tablet orally in a randomised double blind balanced design on the basis of two four-by-four Latin squares. They arrived fasting, and nothing except water to quench thirst was allowed until two hours after dosing, when a light meal was provided.Pupil diameter, radial pulse rate, blood pressure (sitting), intraocular pressure, and prolactin level were measured in that order, before and at 1, 2, 3, and 4 hours after the subjec.t took the tablet. Pupil Correspondence to Dr Q A Mekki. diameter was measured by a pupil gauge,7 blood pressure by a Hawksley random zero sphygmo manometer, intraocular pressure by non-contact tonometery,89 and serum prolactin levels by radioimmunoassay.Volunteers were asked to report any subjective adverse effects. Diazepam and procyclidine were available to treat dystonic reactions if they occurred.The IOP, pupil diameter, heart rate, and blood pressure readings were analysed by calculating the area under the response versus time curve for each subject and on each treatment. These were then analysed by multiple regression analysis after loge transformation to normalise the distribution of the residuals. The areas under the curves for serum prolactin calculated as above were not normally distributed and were therefore analysed by Freidman's two-way analysis of variance.
