Broad-Spectrum Antiviral Activity of an Ankyrin Repeat Protein on Viral Assembly against Chimeric NL4-3 Viruses Carrying Gag/PR Derived from Circulating Strains among Northern Thai Patients

Abstract: Certain proteins have demonstrated proficient human immunodeficiency virus (HIV-1) life cycle disturbance. Recently, the ankyrin repeat protein targeting the HIV-1 capsid, AnkGAG1D4, showed a negative effect on the viral assembly of the HIV-1NL4-3 laboratory strain. To extend its potential for future clinical application, the activity of AnkGAG1D4 in the inhibition of other HIV-1 circulating strains was evaluated. Chimeric NL4-3 viruses carrying patient-derived Gag/PR-coding regions were generated from 131 ant… Show more

“…Previously, our group also reported that protein scaffolds including ankyrin repeat protein, zinc finger protein, and αRep protein could be used as intracellular antiviral molecules, and also demonstrated their therapeutic potential as HIV-1 inhibitors [17,[28][29][30][31].…”

“…SupT1 cells that stably express an artificial ankyrin repeat molecule (AnkGAG1D4) binding to the human immunodeficiency virus (HIV) Gag polyprotein showed a reduced permissiveness to HIV-1 infection [ 34 ]. Subsequent studies investigated the molecular mechanisms whereby AnkGAG1D4 inhibited HIV replication, showing that the intracellular expression of AnkGAG1D4 affected the late stages of virus production [ 66 , 67 , 68 , 69 ]. Despite this as a promising strategy, intracellular DARPin delivery may limit in vivo administration of the molecule.…”

“…Certain proteins are currently attracting attention for the interference of HIV-1 replication. The potential of designed ankyrin-repeat proteins (DARPins) has been proposed as an alternative treatment to monoclonal antibodies [ 1 , 2 , 3 ]. Ank GAG 1D4 is an intracellular anti-HIV-1 DARPin that specifically targets HIV-1 capsid CAp24 [ 4 ].…”
