Breakage of CRISPR/Cas9-Induced Chromosome Bridges in Mitotic Cells

Abstract: Chromosomal instability, the most frequent form of plasticity in cancer cells, often proceeds through the formation of chromosome bridges. Despite the importance of these bridges in tumor initiation and progression, debate remains over how and when they are resolved. In this study, we investigated the behavior and properties of chromosome bridges to gain insight into the potential mechanisms underlying bridge-induced genome instability. We report that bridges may break during mitosis or may remain unbroken unt… Show more

“…There are currently numerous studies underway in which STRIDE is implemented to report on the efficiency of different DDR inhibitors. To date, the technique has been used to study events related to USP1 or PARP7 inhibition and formation of chromosome bridges ( Gozgit et al, 2021 ; Rodriguez-Muñoz et al, 2021 ; Simoneau et al, 2022 ). Preliminary validation of the assay in FFPE tissue sections and the already proven applicability of the PLA methodology in human tissues ( Hegazy et al, 2020 ; Lindskog et al, 2020 ) give hope that this technology may be successfully used in further stages of the drug development process.…”

“…Both anaphase bridges and UFBs are resolved or cleaved at the exit of mitosis. Unresolved or non-cleaved bridges are detected as interphase chromosome bridges formed between interphase nuclei ( 11 ).…”
