Bre Enhances Osteoblastic Differentiation by Promoting the Mdm2-Mediated Degradation of p53

Abstract: Bre is a conserved cellular protein expressed in various tissues. Its major function includes DNA damage repair and anti-apoptosis. Recent studies indicate that Bre is potentially involved in stem cell differentiation although pathophysiological significance along with the molecular mechanisms is still unclear. Here, we report that Bre protein was substantially expressed in the bone tissue and its expression was highly upregulated during the osteogenic differentiation. To test a hypothesis that Bre plays funct… Show more

“…Previous studies suggest that upregulation of MDM2 is associated with tumor initiation and metastasis of GCT [ 15 ]. MDM2 gene amplification in osteosarcoma is often associated with p53 inactivation [ 16 ] or osteosarcoma of metastasis and recurrence; we thus propose that MDM2 is closely related to GCT recurrence and may contribute to tumor cell proliferation. Evidence shows that IGF1 promotes tumor cell differentiation and growth through autocrine or paracrine means [ 17 ].…”

“…Anesthesia-induced cognitive impairment is considered to be the most common type of postoperative cognitive impairment. Previous studies have demonstrated that extended exposure to anesthetics resulted in excessive apoptosis of neurons and decreased the self-renewal capacity of neural stem cells, which consequently led to learning and memory deficits in animals ( 37 41 ). miRNA dysregulation has been previously associated with neurodegeneration induced by anesthesia ( 6 , 15 , 24 ).…”

“…HaCaT cells (CRL-2310), HepG2 cells (HB-8065), HFF cells, and HeLa cells (CCL-2) were obtained from the American Type Culture Collection (ATCC; Manassas, VA, USA) and cultured in DMEM supplemented with 10% fetal calf serum. Human 293T cells were cultured according to the description in our prior study [5,47]. All cells were incubated at 37°C in a humidified atmosphere with 5% CO2, and the medium was replaced every 2 days.…”
