BRD7 mediates hyperglycaemia‐induced myocardial apoptosis via endoplasmic reticulum stress signalling pathway

Abstract: Bromodomain‐containing protein 7 (BRD7) is a tumour suppressor that is known to regulate many pathological processes including cell growth, apoptosis and cell cycle. Endoplasmic reticulum (ER) stress‐induced apoptosis plays a key role in diabetic cardiomyopathy (DCM). However, the molecular mechanism of hyperglycaemia‐induced myocardial apoptosis is still unclear. We intended to determine the role of BRD7 in high glucose (HG)‐induced apoptosis of cardiomyocytes. In vivo, we established a type 1 diabetic rat mo… Show more

“…An increase in cardiomyocyte loss and hypertrophy underlies cardiac fibrosis and dysfunction in diabetes . As expected, the Bax/Bcl‐2 ratio, which reflects the rate of apoptosis, was higher in diabetic mice and in HG‐induced cardiomyocytes, consistent with previous findings . Ang II triggers apoptosis and exacerbates cell growth, both of which are vital pathological features in DCM .…”

“…Western blots were performed using previously described methods . The blots were incubated with the following specific primary antibodies overnight at 4°C on a shaker: anti‐C/EBPβ (Abcam, Cambridge, MA, USA and Santa Cruz Biotechnology, Santa Cruz, CA, USA), anti‐ACE2, anti‐ACE, anti‐AT1R, anti‐AT2R, anti‐TGFβ1, anti‐MMP2, anti‐MMP9, anti‐collagen I, anti‐collagen III (all from Abcam), and B‐cell lymphoma/leukaemia‐2 (Bcl‐2), anti‐Bcl2‐associated X protein (Bax) and anti‐MasR (all Cell Signalling Technology, Boston, MA, USA).…”

“…TUNEL assay was used at the cell apoptosis assay. 19 For DNA fragmentation, TUNEL assays kit (Millipore, USA) was used to treat the H9c2 cardiomyoblasts or tissue sections according to manufacturer's instructions. Briefly, cells were fixed with 4% paraformaldehyde for 10 min at room temperature and then washed with PBS twice.…”
