Brain neurochemical and hemodynamic findings in the NY1DD mouse model of mild sickle cell disease

Abstract: To characterize the cerebral profile associated with sickle cell disease (SCD), we used in vivo proton MRI and MRS to quantify hemodynamics and neurochemicals in the thalamus of NY1DD mice, a mild model of SCD, and compared them with wild-type (WT) control mice. Compared with WT mice, NY1DD mice at steady state had elevated cerebral blood flow (CBF) and concentrations of N-acetylaspartate (NAA), glutamate (Glu), alanine, total creatine and N-acetylaspartylglutamate. Concentrations of glutathione (GSH) at stead… Show more

“…Taurine, also known as β ‐aminobutyric acid, is a kind of sulfur‐containing nonprotein amino acid. Taurine can protect the antioxidant activity of blood cells and prevent the tissue from oxidation and free radicals (Cui et al, ). The down‐regulation of phenylalanine, tyrosine and trytophan as well as the up‐regulation of arginine, praline and taurine in the GFC‐treated group implied that GFC had significant therapeutic effects on PD by regulating amino acid metabolism.…”

“…Pearson's correlation coefficient mapping was carried out on the HbO data (any data likely to include the scalp and skull was removed) to determine the extent of activation (Huppert et al, 2006;Cui et al, 2017). The correlation map of the brain was generated by using the time trace of the CL-HP SS ROI within the activated region and then calculating the time correlation coefficient of this trace to all other voxels in the brain.…”

“…All MRI and 1 H MRS data were acquired in a 9.4-T Varian Direct Drive system (Agilent Technologies, Inc., Santa Clara, CA) (Cui et al 2017). A 14-mm-diameter receive-only surface RF coil (Doty Scientific Inc., Columbia, SC) along with a 7-cm ID 1 H transmit and receive body coil (M2M Imaging Co., Cleveland, OH) was used.…”

“…1 H MRS and brain metabolite quantification 1 H MRS (Cui et al 2017) was acquired via a LASER sequence (Garwood and DelaBarre 2001) with a 1.1 × 2.7 × 1.6-mm 3 voxel positioned in the hippocampus. 1 H MRS acquisition parameters were as follows: time of repetition (TR) = 3.5 s, time of echo (TE) = 36 ms, transients (NT) = 512, complex points = 1024, and spectral width (SW) = 4006 Hz.…”
