Brain Microglial Cells Are Highly Susceptible to HIV-1 Infection and Spread

Abstract: Macrophages are a target of human immunodeficiency virus type 1 (HIV-1) infection and may serve as an important reservoir of the virus in the body, particularly after depletion of CD4 T cells in HIV/AIDS. Recently, sterile alpha motif and histidine/aspartic acid domain-containing protein 1 (SAMHD1) was identified as the major restriction factor of HIV-1 infection in myeloid cells. SAMHD1 is targeted for proteolytic degradation by Vpx, a viral protein encoded by HIV-2 and many simian immunodeficiency viruses bu… Show more

“…Microglia are the primary target for HIV infection in the CNS (Lee et al, 1993;Cenker et al, 2017). Microglia can remove excess and dysfunctional synapses by phagocytosis during neural circuit formation and in specific disease conditions (Stevens et al, 2007;Kettenmann et al, 2013;Brown and Neher, 2014;Xavier et al, 2014;Hong et al, 2016b;Vasek et al, 2016).…”

“…In contrast, fibrinogen could be observed in more blood vessels of adults infected with SIV (b, as overlap of red and green staining). The proportion of vessels with measurable leakage of fibrinogen was significantly higher in adults than infants (c) infection with HIV (Cenker et al 2017) and may become viral reservoirs (Castellano et al 2017). In late-stage disease, microglia can express CD163 and upregulate IBA-1, both classical markers for macrophages (Borda et al 2008;Bortell et al 2018).…”

“…So what causes microglial dysfunction/loss in the setting of very high CSF VL? Microglia and macrophages are known to be infected by HIV and SIV (18)(19)(20) and have been proposed to act as reservoirs for the virus (21,22). Recently, however, it has been proposed that acute infection of human microglia and macrophages instead results in apoptosis, with only a small population of those cells eventually surviving the infection and silencing viral replication, thus becoming viral reservoirs (21).…”
