Introduction: Lead is the most ubiquitous hazardous toxin in the environment. It’s a severe threat to public health and especially to the male reproductive system. In recent years, the usage of the antioxidant to reduce the toxicity of heavy metals has expanded globally. Antioxidants can prevent or minimise the oxidation of other molecules by ROS in a tissue or cell. Grape seeds are natural, rich sources of antioxidant compounds. Aim/Objective: The present study was undertaken to investigate the effect of GSE (grape seed extract) against lead acetate-induced testicular oxidative damage on testis of Wistar rats. Materials and Methods: 24 male Wistar rats were used in the study. They were split into two groups: Group I was the control group (6 rats), and Group II had 18 rats that were given LA at 50 mg/kg BW for 28 days. On the 28th day, all 18 rats were subdivided into three groups: Group II(a) (LA Cessation), Group II(b), and Group II(c) treated with GSE once a day, orally, up to the 56th day. A preliminary phytochemical analysis was conducted. The testicular weight, enzymatic and non- enzymatic oxidative stress markers were estimated. Results: The phytochemical screening showed that bioactive compounds present in the GSE, including phenols, tannins, flavonoids, anthocyanin, glycosides, triterpenoids, and alkaloids. In addition, there was a significant decrease in SOD, CAT, GPx, and GSH levels and a significantly elevated MDA concentration. However, post-treatment with GSE significantly restored the testicular oxidative damage caused by LA in the testis. Conclusion: We concluded that the GSE may have the potential to provide a promising therapeutic effect against LA-induced testicular toxicity.

The present study has aimed to examine the effects of olive leaf extract on the antioxidant enzyme activity of the invertebrate model organism Galleria mellonella L. (Lepidoptera: Pyralidae) hemolymph. For this purpose, after the olive leaves collected from Çanakkale province were dried at room temperature, the ethanolic extract was obtained with a soxhlet. After the ethanol was removed from the obtained extract in the rotary evaporator, the obtained dry matter was prepared in the phosphate buffer salt, 72 mg mL-1 as the highest dose and the lowest 0.010 mg mL-1 and injected into the larvae reared for the experiment. Hemolymph was collected at 4, 8, 12, 16, and 24-hours post-injection. Catalase (CAT) and superoxide dismutase (SOD) enzyme activities and malondialdehyde (MDA) formation levels were determined spectrophotometrically in the collected hemolymph. According to the results obtained, olive leaf extract decreased CAT and SOD activity in G. mellonella hemolymph at certain doses and increased the amount of MDA. For all enzymes, group, dose, and group-dose interactions were statistically significant. In addition, as a result of the examination made in terms of time, it was seen that the measurement of enzyme activities at the 24th hour was significant. The results of our study show that the antioxidant capacity of the olive leaf cannot be transferred to other living things and even reduces the antioxidant capacity of other organisms.

Penile transplantation using vascularized composite allografts is an emerging technique to treat genital loss. In the United States, this procedure has been performed successfully at Massachusetts General Hospital in a patient who had previously undergone treatment for penile cancer. The Johns Hopkins Medical Institutions has developed a research protocol to perform penile transplantation in patients with genital loss secondary to trauma. The process of selecting the appropriate candidate for genitourinary (GU) vascularized composite allograft surgery is rigorous including extensive physical examination, laboratory testing, imaging and psychological evaluations. After transplantation, limiting the potential complications associated with immunosuppression is critical given that the procedure is intended to improve quality of life and is not life-saving. Ultimately, penile transplant is a surgical intervention which may have numerous applications. Optimization of the pre-operative screening process, surgical technique, and immunosuppressive protocol is required to establish this method as the standard treatment for patients with genital loss and limited reconstructive options.
