Brain derived neurotrophic factor expression and DNA methylation in response to subchronic valproic acid and/or aldosterone treatment

Abstract: Aim To test the hypothesis that valproic acid treatment positively affects brain-derived neurotrophic factor (BDNF) expression and DNA methylation in the hippocampus and brain cortex of rats simultaneously treated with aldosterone. Methods Male Sprague-Dawley rats (N = 40) were treated for two weeks with valproic acid (100 mg/1 kg body weight/day) in drinking water and aldosterone (2 μg/100 g body weight/day) or placebo via subcutaneous osmotic minipumps. … Show more

“…In an animal model of SCI and under conditions of cellular excito- toxicity, VPA functions via HDAC inhibition, which increases cell viability, enhances BDNF expression, prevents the activation of NF-kB, and inhibits ER stress (Scheuing et al, 2014). However, there are some studies describing an actual decrease in the gene expression levels of BDNF in the brain after VPA administration (Umka et al, 2010;Buzgoova et al, 2019). It has been well-documented that VPA performs an important role in the treatment of CNS damage, via the induction of neuronal differentiation and the enhancement of neural growth (Kim et al, 2005).…”

“…In conclusion, the present study provides evidence on antidepressant effects of valproic acid using a classical behavioral approach for testing the efficacy of antidepressant drug in animal models. It may be related to the epigenetic modulations induced by the same dose of valproic acid described recently (Buzgoova et al 2019).…”

“…In the case of small intestine, its content was removed before tissue homogenisation. Total RNA extraction, transcription of mRNA into cDNA as well as gene expression quantification was performed as described previously (Buzgoova et al, 2019). Primer BLAST NCBI software was used to design primers specific for the studied genes as well as reference genes (Tab.…”
