Brain Aging, Cognition in Youth and Old Age and Vascular Disease in the Lothian Birth Cohort 1936: Rationale, Design and Methodology of the Imaging Protocol

Abstract: Final outcomes include associations between early and late life cognition and integrity of key white matter tracts, volume of gray and white matter, myelination, brain water content, and visible abnormalities such as white matter lesions and mineral deposits; and influences of vascular risk factors, diet, environment, social metrics, education and genetics on healthy brain aging. It is intended that this information will help to inform and develop strategies for successful cognitive aging.

“…Full brain MRI acquisition parameters have been described previously [Wardlaw et al, 2011]. Briefly, all subjects had brain MRI using the same 1.5 Tesla GE Signa Horizon HDx clinical scanner (General Electric, Milwaukee, WI, USA) at Waves 2 and 3 (73 and 76 years); mean time between scanning was 3.7 years (SD = 0.26, range = 2.2–4.8).…”

“…Briefly, all sequences were co‐registered and tissue volumes quantified using Gaussian mixture modeling cluster analysis [Valdés Hernández et al, 2010; Wardlaw et al, 2011]. According to Standards for Reporting Vascular changes on nEuroimaging (STRIVE), we explicitly defined WMH as punctate, focal, or diffuse lesions in all subcortical regions [Wardlaw et al, 2013].…”

“…These individuals were first tested at 11 years of age in 1947 using a general intelligence test and since the age of 70 have been cognitively re-evaluated every 3 years. In addition to this longitudinal data on cognition, the LBC1936 study has accumulated an extensive database on genetics, biomedical, social and lifestyle factors, and longitudinal brain imaging, resulting in a highly characterised cohort [6] [7] [8] [9]. Post-mortem brains have been donated by 9 nondemented individuals, and to date, there is pre-mortem authorisation for brain donation from 173 individuals.…”

“…In addition to standard structural T2‐, T2*‐, and FLAIR‐weighted MRI, the imaging protocol included a high‐resolution T1‐weighted volume sequence acquired in the coronal plane with field‐of‐view of 256 × 256 mm, imaging matrix 192 × 192 (zero‐filled to 256 × 256), 160 1·3‐mm thick slices giving 1 × 1 × 1·3‐mm voxel dimensions 13. The repetition, echo, and inversion times were 10, 4, and 500 ms respectively.…”
