Bradykinin-induced in vitro contraction of rat mesangial cells via a B2 receptor type

Abstract: The effect of bradykinin (BK) on the contraction of rat mesangial cells (MC) was compared with that of various vasoactive agents. BK induced a dose-dependent contraction [one-half maximal effective dose (ED50) = 50 nM] inhibited by the B2 antagonist, HOE-140 (ED50 = 10 nM). BK-induced MC contraction was independent of extracellular calcium and was reduced by inhibition of protein kinase C (PKC). Neomycin completely prevented the increase in intracellular calcium and the formation of inositol 1,4,5-trisphosphat… Show more

“…A comparable bradykinininduced decrease in pre-and postglomerular arteriolar resistance has, however, been found in rats [18] and dogs [19]. Also it cannot be excluded that blockade of the bradykinininduced mesangial cell contraction contributed to the rise in FF in our model [20].…”

“…For immunoprecipitation of the receptor we applied antiserum AS346 which has been raised against a peptide derived from the COOH terminus of the B 2 wt (positions 329 to 364) (21). In 35 S labeling experiments (Fig. 4d), immunoprecipitation of B 2 wt resulted in a diffuse band of 60 -100 kDa that was superimposable with the immunoreactive band found in 32 P labeling experiments (Fig.…”

“…Mesangial cells are potential mediators of the glomerular damage that we report in this article because they express the B2 receptor and respond in various ways to bradykinin (17,18). However, the increase in interstitial fibrosis that we find in the diabetic mice lacking the B2 receptor suggests that the increased nephropathy in the double mutants must involve events additional to those mediated by mesangial cells.…”
