Bovine babesiosis: induction of protective immunity with culture-derived Babesia bovis and Babesia bigemina immunogens

Abstract: The comparative protective efficacy of culture-derived Babesia bovis and B. bigemina immunogens against challenge exposure of susceptible crossbred cattle to heterologous strains was examined and correlated with the antigenic threshold requirements for induction of protective immunity. Strong protection was observed in animals vaccinated with 10 ml-equivalent doses of soluble, B. bovis exoantigen-containing supernatant fluids. Similar protective responses to B. bigemina exoantigens were evident even at 1 ml-eq… Show more

“…Up to 50% mortality in vaccinates has been associated with heterologous strains of Australian B. bovis (Timms et al 1983(Timms et al , 1984. “…Previous vaccination studies have used immuno¯uo-rescence (Kuttler et al 1983) or enzyme immunoassays (Timms et al 1983(Timms et al , 1984Montenegro-James et al 1987) to evaluate the antibody response of vaccinates. Both tests are qualitative in that they detect an antibody response to one or more proteins while not quantifying the degree of antibody response to speci®c immunogens or the number of proteins that have elicited a response.…”

“…Cross-reactive bands were consequently excluded from the evaluation of Western blots with field sera. Previous work describes varying diagnostic antigen patterns for the different Babesia species (Montenegro-James et al 1987; , 1990Hines et al 1989;Gorenflot et al 1991;McElwain et al 1991;Madruga et al 1996, Machado et al 1997Passos et al 1998), which may be due differences in strain composition or preparation protocols. Standard protocols for routine diagnosis must consequently include the definition and propagation of strains and standardised antigen preparation to ensure sufficient reproducibility.…”

“…Since the development of cell culture systems for the continuous propagation of B. bovis parasites (Erp et al 1980;Levy and Ristic 1980), exoantigens derived from cultured parasites have been used for immunizing animals against babesiosis (Kuttler et al 1982(Kuttler et al , 1983Montenegro-James et al 1985, 1987Patatarroyo et al 1995;Zhao and Liu 1991).…”
