Babesia bovis merozoite proteins presenting as exoantigens in in vitro culture supernatants have been characterized. Bovine antisera to B. bovis exoantigens were used to immunoprecipitate [35S]-methionine metabolically labeled or lactoperoxidase-catalyzed radioiodinated B. bovis merozoite proteins. A total of 24 metabolically labeled proteins ranging in molecular weight from 24,000 to 225,000 Da and 9 radioiodinated proteins with molecular weights varying between 24,000 and 225,000 Da were identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Monoclonal antibodies to B. bovis merozoite surface proteins were also used to immunoprecipitate metabolically labeled exoantigens directly from in vitro culture supernatants. These results demonstrate epitopes from at least nine merozoite surface proteins present in the exoantigen fraction, among which are the recently characterized major surface antigens 1 and 2, rhoptry-associated protein 1, and spherical body protein 2.

To establish an assay for the serological differentiation of bovine Babesia species ( B. bigemina, B. bovis, B. divergens and B. major), antigens from experimentally infected cattle were Western blotted and probed with homologous and heterologous sera. Varying antigen patterns for each species allowed the determination of species-specific diagnostic antigens. Blood samples from 36 naturally infected cattle from the province of Styria were tested by indirect immunofluorescence antibody test (IFAT) against B. divergens, as well as by Western blotting against B. bigemina, B. bovis, B. divergens and B. major, 3 weeks after clinical babesiosis was diagnosed by blood smears. All 36 cattle were B. divergens-positive when tested by IFAT. In four cases (11%), an infection with both B. bovis and B. divergens and in two cases a single infection with B. bovis were diagnosed when tested by Western blot. B. bigemina and B. major infections were not detected. These are the first serologically confirmed cases of B. bovis in Austrian cattle.

Culture-derived Babesia orientalis exoantigens used as a vaccine against buffalo babesiosis

Buffalo babesiosis represents a major problem for the livestock industry in China. The pathogen of this disease was isolated and identified as Babesia orientalis and subsequently propagated in vitro, using the microaerophilus stationary phase culture system. The aim of this study was to determine the efficacy of exoantigens derived from B. orientalis cultures to induce a protective immunity against challenge exposure to virulent organisms in laboratory and field. The results showed that exoantigens of B. orientalis could induce a high degree of protection against challenge with a virulent B. orientalis strain. The animals in the vaccinated group exhibited a slight decrease in haemoglobin levels and blood cell counts, whereas animals in the control group showed typical clinical symptoms and died between days 11 and 16 after challenge. In another approach, 82 buffaloes kept in B. orientalis-endemic areas were injected with exoantigens plus Freund's adjuvant in two doses at an interval of 2 weeks, one month before the endemic period. An additional 86 buffaloes served as unvaccinated controls. During the endemic period, the clinical signs of all buffaloes were monitored and the antibody response was investigated, using the latex agglutination test. During the endemic period, none of the vaccinated buffaloes showed clinical signs or died, whereas five control buffaloes showed clinical signs and two of them died. The mean antibody titre of vaccinated animals was higher than that of the control animals. These results showed that the culture-derived exoantigens of B. orientalis could be used as a vaccine to prevent buffalo babesiosis.
