ABSTRACT. Surfactant protein D (SP-D) is a pattern recognition molecule that has an important role in pulmonary host defense. In this study, we developed an enzyme-linked immunosorbent assay (ELISA) for bovine SP-D and determined the concentration of SP-D in bronchoalveolar lavage fluid (BALF) from calves. Bovine SP-D was purified from BALF using a mannose-Shepharose 6B column. The obtained 44 kDa protein was identified as bovine SP-D by N-terminal amino acid sequence analysis and SDS-PAGE analysis. The peptides corresponding to bovine SP-D amino acid residues SDTRKEGT, which have little homology across bovine serum collectins, were synthesized and used to raise an antibody in rabbits. The obtained antibody was specific for bovine SP-D and did not react with collectins in serum. The anti-bovine SP-D antibody was purified and an ELISA system was developed. The detection range of this assay was 4-125 ng/ml, and the intra-assay and inter-assay coefficients of variation were 5.6 and 9.7%, respectively. The concentrations of SP-D in BALF collected from calves experimentally infected with bovine adenovirus type-3 or Mannheimia haemolytica were determined by the ELISA. Elevation of SP-D was found in BALF from inoculated lobes of infected calves compared with those of non-inoculated lobes and those from control animals. These data suggest that the ELISA developed in this study may be available to investigate the physiological role of bovine SP-D in bovine lung. Surfactant protein D (SP-D) is a protein component of pulmonary surfactant that is synthesized and secreted by alveolar type II cells [5]. SP-D is a member of the collectin subgroup of the C-type lectin superfamily [2], along with surfactant protein A (SP-A) in lung and mannose-binding lectin (MBL), conglutinin, and CL-43, which are present in serum [8]. These collectins have a common molecular structure that consists of 4 distinct domains: an amino terminus containing a disulfide bond, a collagen-like domain consisting of Gly-X-Y repeats, a neck region, and a carbohydrate recognition domain (CRD). The oligometric structure of SP-D consists of a cruciform dodecamer of four trimeric subunits, each of which is composed of a 43 kDa monomer [26].Pulmonary surfactant, which is a mixture of lipids and protein, covers the peripheral airway and protects against invasion by microorganisms as the first line of host defense. SP-D has a role in the regulation of surfactant lipid metabolism [14]; however, it has been emphasized that SP-D is an important molecule in innate immune defense, like the other collectins [18,26]. SP-D binds or agglutinates a broad range of pathogenic microorganisms, including bacteria, viruses, fungi, and yeasts, mediated by the CRD regions in the molecule [18] [6,7,22,25,27]. Deer mastadenovirus B pneumonia in a white-tailed deer fawn

A 7-mo-old farmed white-tailed deer fawn ( Odocoileus virginianus) died after several weeks of progressive deterioration associated with endoparasitism and respiratory signs. A field autopsy was performed, and lung tissue was submitted for histologic examination. The findings were consistent with necrosuppurative bronchointerstitial pneumonia with intranuclear viral inclusions. Immunofluorescence using fluorescently labeled polyclonal antibodies to bovine adenovirus 3 and 5 was positive. To rule out cross-reactivity with other adenoviruses, formalin-fixed, paraffin-embedded tissue sections were submitted for genome sequence analysis, which revealed a 99.6% match to Deer mastadenovirus B (formerly Odocoileus adenovirus 2, OdAdV2). To our knowledge, natural clinical disease associated with OdAdV2 has not been reported previously.

Diagnostic tests to identify pathogens involved in respiratory diseases of cattle are increasingly used, predominantly driven by the need to rationalize antimicrobial use. Several methods to sample the respiratory tract are available, of which a deep nasopharyngeal swab, transtracheal wash, and nonendoscopic bronchoalveolar lavage best fit practice. Each technique has advantages and disadvantages, and consensus regarding the best choice is not yet reached. Next to microbial culture, for microbiologic diagnosis polymerase chain reaction is especially popular. Promising techniques for a rapid diagnosis are matrix-assisted laser desorption/ionization time of flight mass spectrometry and next-generation sequencing, which will become widely available in coming years. It is still difficult to interpret identification of opportunistic pathogens, both at the individual and herd level. More research is needed before evidence-based guidelines can be developed.
