The focus of the present work is to shed light on possible modifications of the molecular properties of polysaccharide hydrogels induced by the establishment of specific non-covalent interactions during the loading of a guest compound inside the gel phase. With this aim, a case study of the encapsulation of caffeine (Caf) inside cyclodextrin-based hydrogels, namely, cyclodextrin nanosponges (NS), is systematically investigated here by using UV Raman scattering experiments. The UV Raman spectra of the hydrogels, analysed as a function of temperature, concentration of the guest molecule loaded in the gel phase and pH, prove particularly informative both on the structural rearrangements of the hydrophobic/hydrophilic groups of the polymeric network and on the breaking/formation of specific guest-matrix interactions.Analysis of the temperature dependence of dynamical parameters, i.e., the dephasing time associated with specific vibrational modes of the polymer backbone, enables the proposal of a molecular picture in which the loading of Caf in NS hydrogels tends to favour access of the water solvent to the more hydrophobic portions of the polymer matrix, which is in turn reflected in a marked increase in the solvation of the whole system. The achievements of this work appear of interest with respect to the design of new possible strategies for controlling the diffusion/release of bioactive molecules inside hydrogel networks, besides corroborating the potential of UV Raman scattering experiments to give new molecular insights into complex phenomena affecting hydrogel phases. A IntroductionHydrogels are a class of soft materials that have attracted in recent years great interest from chemical and physical scientists. 1 Hydrogels are three-dimensional structures with the capability to retain large amounts of water while maintaining their elastic texture. 2,3 In the case of polymeric hydrogels, chemically or physically cross-linked networks of polymer chains create a characteristic three-dimensional matrix with interstitial spaces that are capable of hosting water. 4 The unique mechanical and physical properties of hydrogels, together with their biocompatibility and biodegradability, have made these materials attractive for a wide range of technological applications. For example, the physiochemical similarity of hydrogels to the native extracellular matrix suggested the use of these materials as scaffolds for tissue engineering and regenerative medicine. 5-7Indeed, synthetic matrices where cells can grow and interact with the environment in all three dimensions allow the in vivo behaviour of cells to be mimicked more efficiently than in previous methods based on the 2D growth of cells. In this work, the clinical anticellulite efficacy of a commercial extract of Trichilia catigua (Catuaba) and Ptychopetalum olacoides Bentham (Marapuama) was evaluated. Twenty-seven woman subjects between the ages of 20 and 40 years old with moderate gynoid lipodystrophy participated in this study. The Trichilia catigua and Ptychopetalum olacoides Bentham extract was incorporated into a cosmetic emulsion, and the subjects applied this formulation once a day for 60 days. The following parameters were assessed: hypodermic layer thickness by B-mode ultrasonography, cutaneous temperature, and anthropometric measurements. The test product caused an increase in skin temperature after 60 days application without affecting the hypodermic layer thickness. In addition, a statistically significant reduction in circumference of specific body areas was observed (stomach, waist, upper thigh, upper leg, and upper hip). Thus, the results indicated that the Trichilia catigua and Ptychopetalum olacoides Bentham extract was effective in the reduction of the gynoid lipodystrophy aspect, likely by reducing edema and improving local microcirculation. Neste trabalho, avaliou-se a eficácia clínica anticelulite de um extrato comercial de Trichilia catigua (Catuaba) e Ptychopetalum olacoides Bentham (Marapuama). Participaram deste estudo 27 mulheres voluntárias entre 20 e 40 anos com hidrolipodistrofia ginoide moderada. O extrato de Trichilia catigua e Ptychopetalum olacoides Bentham foi incorporado em uma emulsão cosmética e os voluntários aplicaram esta formulação uma vez por dia, durante 60 dias. Foram avaliados os seguintes parâmetros: espessura da camada hipodérmica por ultrassonografia em modo B, temperatura cutânea e medidas antropométricas. O produto testado propiciou aumento na temperatura da pele após 60 dias de aplicação sem afetar a espessura da camada hipodérmica. Além disso, observou-se uma redução estaticamente significativa na circunferência de áreas corporais específicas (estômago, cintura, parte superior da coxa, parte superior da perna e quadril superior). The solvent‐free encapsulation of caffeine and kojic acid was carried out in different carboxylate‐based MOFs [MIL‐53(Al), UiO‐66 and Mg‐MOF‐74] by high pressure (0.32 GPa) contact. This methodology enables fast and ecofriendly encapsulation and gives rise to additive@MOFs with physical and features equivalent to those of materials obtained by common liquid phase encapsulation processes. It could be applied to other host–guest systems, simplifying the procedures, reducing the use and waste of harmful chemicals, and approaching the conditions of interest in the industry. The characterization carried out by thermogravimetry, X‐ray diffraction, N2 adsorption, and 13C NMR provided information about the presence and conformation of the additives in the MOFs. The highest encapsulation values for caffeine (37 %) and kojic acid (32 %) were obtained with MIL‐53(Al).
