Bone marrow-derived stem/stromal cells (BMSC) 3D microtissues cultured in BMP-2 supplemented osteogenic induction medium are prone to adipogenesis

Abstract: Bone marrow-derived mesenchymal stem/stromal cells (BMSC) may facilitate bone repair through secretion of factors that stimulate endogenous repair processes or through direct contribution to new bone through differentiation into osteoblast-like cells. BMSC microtissue culture and differentiation has been widely explored recently, with high-throughput platforms making large-scale manufacture of microtissues increasingly feasible. Bone-like BMSC microtissues could offer an elegant method to enhance bone repair, … Show more

“…1a, the expression of miR-19b-3p was obviously elevated in osteoporosis group as compared with healthy control group (P < 0.05). To explore the potential role of miR-19b-3p during osteoblast differentiation, the expression of miR-19b-3p was measured in BMSC stimulated with BMP-2, which has been proved to induce osteoblast differentiation [13]. The results indicated miR-19b-3p was significantly increased in BMP-2 induced MSCs as compared with control cells.…”

“…Mesenchymal stem cells have been reported to enhance bone regeneration by exerting auto or paracrine effects via the secretion of growth factors or direct differentiation into bone cells ( 30 ). Previous studies have revealed that mesenchymal stem cells are generally used with scaffolds ( 31 , 32 ).…”

“…Bone marrow mesenchymal stem cells may enhance bone regeneration through secretion of growth factors or through direct differentiation into bone cells ( 16 ). The results of this study suggest combination therapy using stem cells and BMP-2 with a three-dimensional approach.…”
