Bone grafting: Role of histocompatibility in transplantation

Abstract: The role of histocompatibility matching in bone allografting was studied in two canine bone graft models. In a cancellous ulnar segmental replacement model, frozen bone allografts exchanged between closely matched dogs were significantly better incorporated by radiographic and histologic criteria than were strongly incompatible grafts. Frozen allografts from disparate donors in recipients receiving immunosuppression appeared indistinguishable 6 months later from those in the untreated closely matched groups an… Show more

“…counterparts with regard to bone induction properties, revascularization, and incorporation into host bone (Goldberg et al 1985, Wilson et al 1985, Burchardt 1987. Clinical matching of transplantation antigens between donor and host has not given a definite answer as to whether allograft inferiority is due to immunologic incompatibility (Friedlaender 1983, Burwell et al 1985, although experimental evidence suggests that this is the case (Bos et al 1983, Goldberg et al 1985.…”

Analogous to cell-mediated mechanisms in skin-graft rejection, T-lymphocyte immune responses might be important for the revascularization and incorporation of allogeneic bone grafts.…”

“…Standards of measurement were chosen based on a consensus among three orthopaedic surgeons, who constituted a consensus development panel. As a result, three indices of the Goldberg scoring system were chosen [13]. On lateral foot radiographs, the first index is evaluation of graft appearance, which receives a score of 0 for resorbed, 1 for mostly resorbed, 2 for largely intact, and 3 for reorganizing.…”

“…Fracture healing was analysed by two observers, without knowledge of the treatments, according to the classification of Goldberg et al (Goldberg et al, 1985) with stage 0 indicating radiological non-union, stage 1 indicating possible union and stage 2 indicating radiological union. Callus diameter was measured in relation to the femur diameter (%) (ImageJ Analysis System, NIH, Bethesda, MD, USA).…”
