Bone complications in children with Gaucher disease

Abstract: For paediatric patients with Gaucher disease, enzyme replacement therapy (ERT) has the potential to prevent the development of serious, irreversible skeletal complications. Analysis of skeletal data for paediatric patients receiving ERT must take into account the pubertal growth spurt and developmental changes in bone marrow composition. In a study conducted at the Burlo Garofolo Institute in Trieste, Italy, 10 paediatric patients have received ERT, and data are available for 3-9 years of follow-up. ERT was as… Show more

“…The results from several studies that included children 83,88,113 indicated that during ERT bone thickness and bone density tended to move towards more normal values expected for the age and gender of the child. Rosenthal and colleagues 113 remarked that children in their sample accounted for all of the increase in bone thickness observed.…”

“…(17,18) Importantly, it is not known whether osteopenia in GD1 increases the risk of fractures as it does in postmenopausal women. (19) Imiglucerase therapy improves bone mass and bone mineral density (12,13,(20)(21)(22)(23) and studies have shown an apparent decrease in reports of bone pain and bone crises. (23,24) However, it is not known whether imiglucerase treatment decreases the risk of fractures.…”

Rosenthal and colleagues 113 remarked that children in their 103 of gamma camera images at femora/tibia follow-up 31.6 improved (peripheral) and sternum/pelvis/lumbar (central).…”
