BMP9 stimulates joint regeneration at digit amputation wounds in mice

Abstract: A major goal of regenerative medicine is to stimulate tissue regeneration after traumatic injury. We previously discovered that treating digit amputation wounds with BMP2 in neonatal mice stimulates endochondral ossification to regenerate the stump bone. Here we show that treating the amputation wound with BMP9 stimulates regeneration of a synovial joint that forms an articulation with the stump bone. Regenerated structures include a skeletal element lined with articular cartilage and a synovial cavity, and we… Show more

“…While there were 20 KEGG pathways significantly enriched in X1, only four pathways were enriched in X2 ( Figure 2F and Supplementary Table S5 ). To confirm the results of the bioinformatics analyses, we quantified the expressions of several genes related to tissue regeneration ( Kroehne et al, 2011 ; Tsujioka et al, 2017 ; Galvao et al, 2018 ; Yu et al, 2019 ). prg4a and klf11 were indeed upregulated only in the lesioned hemisphere ( Figure 2G ), while gfap and il6st were so in both hemispheres ( Figure 2H ).…”

“…Recently, it has been reported that ectopic Fgf signaling can induce intercalary regeneration in chicken limb buds 36 . Likewise, mouse digit tip regeneration can be enhanced by Bmp application 37,38 . These related discoveries imply the existence of a conserved regeneration system across species.…”

“…Bone morphogenetic protein 9 (BMP9) (also known as growth differentiation factor 2, or GDF‐2), a member of the transforming growth factor‐β (TGF‐β)/BMP superfamily, is principally produced by the liver and circulates in the bloodstream; its receptors are ALK1 and ALK2 . Recent research has found that BMP9 plays an important role in preventing liver fibrosis, improving heart failure, suppressing tumors, regulating glucose and lipid metabolism, and regenerating joints . More recently, dentinogenetic and tooth‐alveolar bone complex defects in BMP9‐knockout mice were reported, demonstrating the role of BMP9 in the skeletal system.…”
