BMP2/7 heterodimer is a stronger inducer of bone regeneration in peri-implant bone defects model than BMP2 or BMP7 homodimer

Abstract: This study aimed to compare the effects of bone morphogenetic protein BMP2/7 heterodimer and BMP homodimers on bone regeneration in bone defects model. Identical peri-implant bone defects model were created using proper controls on the frontal skull in 18 minipigs. Collagen sponges with low-dose (30 ng/mL) BMP2/7 heterodimer, BMP2 or BMP7 homodimer were filled in the defects. New bone formation and the expression of type I collagen (Col1), alkaline phosphatase (ALP) and osteocalcin (OCN) were evaluated after 2… Show more

“…On the other hand, proteins such as BMP2 have been shown to enhance bone formation on titanium implants (Schliephake et al 2012;Schliephake et al 2008), and BMP2/7 heterodimers have been found to exert osteogenic effects on implant surfaces (Sun et al 2012). In the periimplantitis bone tissue investigated here, BMP7 was downregulated, and BMP2 was slightly upregulated.…”

“…For example, BMP4/7 heterodimer induces stronger activity of mesoderm than BMP4 or BMP7 homodimers in Xenopus. Similarly, BMP2/7 heterodimer has higher activity in bone regeneration than BMP2 or BMP7 homodimers in mammals (46). In addition, BMPs bind to various BMP receptors with different affinities.…”

“…In these previous studies, large segmental bone defects in rats were healed by the implantation of BMP‐2 gene‐activated muscle tissue fragments. Other studies have demonstrated that gene‐enhanced bone repair can be improved by use of growth factor combinations and, recently, the use of BMP heterodimers has emerged as a potent strategy to induce osteogenesis more effectively compared to the homodimer BMP‐2 . However, to date, the osteogenic effect of BMP heterodimers on muscle tissue fragments has not yet been examined and it is unknown whether the muscle‐based expedited gene therapy approach could be improved by use of heterodimers.…”
