Blueberry Supplementation Enhances Signaling and Prevents Behavioral Deficits in an Alzheimer Disease Model

Abstract: Previously, we showed that blueberry (BB) supplementation reversed the deleterious effects of aging on motor behavior and neuronal signaling in senescent rodents. We now report that BB-fed (from 4 months of age) APP + PS1 transgenic mice showed no deficits in Y-maze performance (at 12 months of age) with no alterations in amyloid beta burden. It appeared that the protective mechanisms are derived from BB-induced enhancement of memory-associated neuronal signaling (e.g. extracellular signal-regulated kinase) an… Show more

“…The amount of corn in the control diet was adjusted to compensate for the added volume of the coffee powder or caffeine. The control NIH-31 diet was the same as that used in earlier studies in which other foods were found to be beneficial for mitigating brain aging (Youdim et al 2000;Joseph et al 2003;Casadesus et al 2004;Goyarzu et al 2004;Shukitt-Hale et al 2005). The coffee powder, supplied by Kraft Foods Global, Inc. (Northfield, IL, USA), was prepared by lyophilizing brewed coffee.…”

“…The blueberry was then added to the control diet in place of cornstarch, yielding a chow consisting of control diet plus blueberry at 2% (g per kg diet). The diets used in this study are isocaloric and have been used previously (Joseph et al, 1999(Joseph et al, , 2003Willis et al, 2005); for additional information regarding dietary formulations see Joseph et al, 1999). The transplant recipients were maintained on the respective diets for the duration of the study.…”

“…These models are characterised by enhanced indices of oxidative stress and inflammation along with disruption of the dopaminergic system, similar to that observed in healthy, aged animals (187) . Furthermore, a blueberry-rich diet was also shown to be protective in AD models (amyloid precursor protein/PS1 transgenic mice), preventing spatial memory deficits along with enhancement of memory-associated neuronal signalling (180) . In particular, blueberry-supplemented amyloid precursor protein/PS1 mice exhibited greater levels of hippocampal ERK activation as well as hippocampal PKCa activation, both known to be involved in regulation of synaptic plasticity and consolidation of learning and memory (180) .…”

“…There have been many studies reporting the potential effects of berry supplementation on spatial memory in aged animals (27,(180)(181)(182)(183) . Early studies indicated that long-term supplementation (from 6 to 15 months of age) with berries (blueberry or strawberry) retards age-related decrements in cognitive and neuronal function (181) .…”
