The genus presents high species richness in the Argentine Sea and DA has been frequently detected in the last few years in plankton and shellfish samples, but the species identity of the producers remains unclear. In the present work, the distribution and abundance of Pseudo-nitzschia species and DA were determined from samples collected on two oceanographic cruises carried out through the Argentine Sea ($39-47 S) during summer and spring 2013. Phytoplankton composition was analysed by light and electron microscopy while DA was determined by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). The genus Pseudo-nitzschia was recorded in 71 and 86% of samples collected in summer and spring, respectively, whereas DA was detected in only 42 and 21% of samples, respectively. Microscopic analyses revealed at least five potentially toxic species (P. australis, P. brasiliana, P. fraudulenta, P. pungens, P. turgidula), plus putatively non-toxigenic P. dolorosa, P. lineola, P. turgiduloides and unidentified specimens of the P. pseudodelicatissima complex. The species P. australis showed the highest correlation with DA occurrence (r = 0.55; p < 0.05), suggesting its importance as a major DA producer in the Argentine Sea. In the northern area and during summer, DA was associated with the presence of P. brasiliana, a species recorded for the first time in the Argentine Sea. By contrast, high concentrations of P. fraudulenta, P. pungens and P. turgidula did not correspond with DA occurrence. This study represents the first successful attempt to link toxigenicity with Pseudo-nitzschia diversity and cell abundance in field plankton populations in the south-western Atlantic.

Some members of the diatom genus Pseudo-nitzschia produce the toxin domoic acid (DA), which through trophic transfer causes mass mortalities of wildlife, shellfish harvesting closures, and risks to human health. Nutrient and micronutrient limitation have been shown to regulate DA production. This study tested the hypothesis that changing partial pressure of CO 2 (pCO 2 ) can interact with nutrient limitation to help determine cellular DA levels, an environmentally relevant issue in light of current increases in atmospheric pCO 2 . Cultures of the toxic species Pseudo-nitzschia multiseries were incubated using semicontinuous methods under a matrix of three pCO 2 conditions: , 22 Pa (220 ppm), , 41 Pa (400 ppm), and , 74 Pa (730 ppm), and two phosphate concentrations: 20 mmol L 21 , P-replete; and 0.5 mmol L 21 , P-limited. DA production was regulated by both pCO 2 and phosphate availability. DA concentrations were 30-50 times higher in P-limited cultures compared to Preplete ones, at the same pCO 2 levels. Increasing CO 2 levels stimulated DA production under both nutrient conditions, but especially in P-limited cultures, where DA levels increased approximately four times over the pCO 2 range examined. Growth rates, primary productivity, photosynthesis vs. irradiance parameters, and cellular elemental ratios also responded interactively to the availability of both CO 2 and phosphate. Our results raise the possibility that growth rates and toxicity of the diatom Pseudo-nitzschia multiseries could increase substantially in the future high-CO 2 ocean, suggesting a potentially escalating negative effect of this harmful algal bloom species on the future marine environment.

Ocean processes are dynamic, complex, and occur on multiple spatial and temporal scales. To obtain a synoptic view of such processes, ocean scientists collect data over long time periods. Historically, measurements were continually provided by fixed sensors, e.g., moorings, or gathered from ships. Recently, an increase in the utilization of autonomous underwater vehicles has enabled a more dynamic data acquisition approach. However, we still do not utilize the full capabilities of these vehicles. Here we present algorithms that produce persistent monitoring missions for underwater vehicles by balancing path following accuracy and sampling resolution for a given region of interest, which addresses a pressing need among ocean scientists to efficiently and effectively collect high-value data.More specifically, this paper proposes a path planning algorithm and a speed control algorithm for underwater gliders, which together give informative trajectories for the glider to persistently monitor a patch of ocean. We optimize a cost function that blends two competing factors: maximize the information value along the path, while minimizing deviation from the planned path due to ocean currents. Speed is controlled along the planned path by adjusting the pitch angle of the underwater glider, so that higher resolution samples are collected in areas of higher information value. The resulting paths are closed circuits that can be repeatedly traversed to collect long-term ocean data in dynamic environments. The algorithms were tested during sea trials on an underwater glider operating off the coast of southern California, as well as in Monterey Bay, California. The experimental results show improvements in both data resolution and path reliability compared to previously executed sampling paths used in the respective regions.
