Characterization and Toxicity Analysis of Lab-Created Respirable Coal Mine Dust from the Appalachians and Rocky Mountains Regions

Coal mine workers are continuously exposed to respirable coal mine dust (RCMD) in workplaces, causing severe lung diseases. RCMD characteristics and their relations with dust toxicity need further research to understand the adverse exposure effects to RCMD. The geographic clustering of coal workers’ pneumoconiosis (CWP) suggests that RCMD in the Appalachian region may exhibit more toxicity than other geographic regions such as the Rocky Mountains. This study investigates the RCMD characteristics and toxicity based on geographic location. Dissolution experiments in simulated lung fluids (SLFs) and in vitro responses were conducted to determine the toxicity level of samples collected from five mines in the Rocky Mountains and Appalachian regions. Dust characteristics were investigated using Fourier-transform infrared spectroscopy, scanning electron microscopy, the BET method, total microwave digestion, X-ray diffraction, and X-ray photoelectron spectroscopy. Inductively coupled plasma mass spectrometry was conducted to determine the concentration of metals dissolved in the SLFs. Finer particle sizes and higher mineral and elemental contents were found in samples from the Appalachian regions. Si, Al, Fe, Cu, Sr, and Pb were found in dissolution experiments, but no trends were found indicating higher dissolutions in the Appalachian region. In vitro studies indicated a proinflammatory response in epithelial and macrophage cells, suggesting their possible participation in pneumoconiosis and lung diseases development.

Background: Dental technician expose to large amounts of crystalline silica that increase their risk of developing silicosis. Neopterin are produced upon stimulation with the cytokine interferon-γ. Neopterin is produced by activated monocytes, macrophages, and dendritic cells upon stimulation by interferon gamma produced by Tlymphocytes. Neopterin production provides prognostic information in patients with autoimmune diseases. Objective: Assessment of serum neopterin, inflammatory cytokines, enzyme activities and trace elements in smokers and non smokers dental technicians and to compare them with the corresponding levels in a control group. Subjects and Methods: Serum was examined in 45 dental technician, 15 subjects as a control group and 30 subjects exposed to silica. The surveyed dental technician were divided into three groups: 1 st group (control group), 2 nd group (smokers dental technician) and 3 rd group (non smokers dental technician). Serum neopterin, some trace elements, LDH, ALP, ACP, TNF-α, and IL-8 were determined. Result: Higher levels of neopterin were established in smokers dental technician exposed to crystalline silica (16.48 ng/ml) by comparison with non smokers dental technicians (9.85 ng/ml) and the control group (4.24 ng/ml). Significant increase in the levels of silica, calcium, copper, enzymatic activities, TNF-α, IL8 and a significant decrease in Zn levels in serum of smokers subject compared with both non smokers and control subjects. Conclusion: Research on biomarkers for silicosis is still at its nascent stage. The concentration of serum neopterin could be used as a biomarker in the diagnostic criteria for silicosis moreover it is suitable for introduction into the routine clinical laboratory practice.

Objectives To explore the value of a logistic regression model based on haematological parameters for the early diagnosis of silicosis by comparing the differences in haematological parameters between silicosis patients and healthy physical examiners.Methods A total of 390 individuals, including 195 silicosis patients and 195 normal participants were included in the training cohort. Then, 65 silicosis patients and 65 healthy individuals were enrolled in the validation cohort. Whole blood samples were collected from all participants, and hematological indicator characteristics were determined. Features with statistical significance in the univariate analysis of the training cohort and reported significant features were included in the logistic regression analysis to determine the independent factors influencing the diagnosis of silicosis and to construct a logistic diagnostic model. A receiver operating characteristic (ROC) curve was plotted to evaluate the accuracy of the model in diagnosing silicosis.Results In the training cohort, several hematological indicators were significantly different in silicosis patients, including Hematocrit(HCT), Hemoglobin(HGB), Mean corpuscular volume(MCV), Red Blood Cell Count(RBC), White blood cell count (WBC), Mon#, Mon%, Neu#, Neu%, Red blood cell distribution width coefficient of variation(RDW_CV), C-reactive protein(CRP), Hydroxybutyrate dehydrogenase (HBDH), Lactate dehydrogenase(LDH), Prothrombin time(PT), International normalized ratio(INR), Fibrinogen(FIB), and D-Dimer(DD) levels, all with statistical significance (P < 0.05). The silicosis diagnostic model performed well in the training cohort (Area Under Curve, AUC = 0.943) and had high diagnostic sensitivity (83.1%) and specificity (92.3%). The diagnostic model also effectively distinguished between silicosis patients and the control cohort in the validation cohort (AUC = 0.936).Conclusions This study confirmed that Age, CRP, LDH, Macro%, and INR were independent factors influencing the diagnosis of silicosis, and the logistic regression model based on these indicators could provide a reliable basis for predicting silicosis diagnosis.
