Blood Flow in Single Surface Arterioles and Venules on the Mouse Somatosensory Cortex Measured with Videomicroscopy, Fluorescent Dextrans, Nonoccluding Fluorescent Beads, and Computer-Assisted Image Analysis

Abstract: Summary: Cortical surface vessels were monitored through closed cranial windows with an epifluorescence microscope and SIT or ICCD cameras. Fluorescent dex trans or 1.3 fLm latex beads were injected into the con tralateral jugular vein for plasma labeling and for vascular transits. For close arterial transits, these tracers or phys iological saline were injected into the ipsilateral external carotid artery. A VTTs were calculated from intensity dif ferences of tracers between a branch of the MCA and a vein dra… Show more

“…Techniques for measurement of amniotic fluid flow into the lung are not available; however, fluorescently labeled polystyrene beads were successfully used previously to measure blood flow (Rovainen et al, 1993). This technology was adapted to evaluate amniotic fluid flow into the fetal lung.…”

“…RBC velocity was determined via analysis of the velocity of 1-mdiameter fluorescent polystyrene microspheres (FluoSpheres, yellow/ green; Molecular Probes, Eugene, OR) injected i.v. in 10 l boluses (23). Beads were visualized via epifluorescence as for FITC dextran.…”

“…In contrast to the substantial interconnections within the surface and subsurface networks, the penetrating arterioles that bridge these networks appear to be largely devoid of anastomoses (4,9). This architecture casts the penetrating arterioles as a singular element for the regulation of blood flow to columnar-sized regions of cortex (10)(11)(12). Yet this very same feature is likely to cause pronounced ischemia if the penetrating arterioles are occluded.…”
