Blood and Tissue Selenium Determination by Hydride Generation Atomic Absorption Spectrophotometry

Abstract: The aim of the study was to optimize the method of blood and tissue selenium determination in ruminants, and to implement it in research and diagnostic practice. A method was developed for selenium determination by hydride generation atomic absorption spectrophotometry (HG-AAS) in full blood, blood plasma and serum, the liver, skeletal muscle tissue, the myocardium, and the kidneys after wet mineralization of samples in a closed nitric acid and hydrogen peroxide system and subsequent hydrogen chloride reductio… Show more

“…After removal of nitric acid from the digested sample in the evaporation rotor of the same apparatus, the water solution was mixed with 20% hydrochloric acid. Selenium in the processed sample was measured using HG-AAS (Hydride Generation Atomic Absorption Spectrometry) according to the method described by Pechova et al (2005) in an AAS Solaar M6 (Unicam, Great Britain). Selenium concentrations were expressed in terms of µg/l of whole blood.…”

“…Milk and whole blood concentration of Se was determined after mineralization of samples by hydride technique using Solaar M6 (Unicam, Great Britain) with electrothermically heated cuvette as per methodology designed by Pechová et al (2005). All analyses were performed by the laboratory of the Clinic of Ruminant Diseases of the University of Veterinary and Pharmaceutical Sciences Brno.…”

“…After vaporization and conversion of the mineralizate into water solution, 20% HCl is added. In such a solution, selenium content is determined in AAS Solaar M6 unit by Unicam (Great Britain) using the hydride technique of atomic absorption spectrophotometry (Pechova et al, 2005).…”
