Blockage of interleukin-6 receptor ameliorates joint disease in murine collagen-induced arthritis

Abstract: Objective. To clarify the role of interleukin-6 (IL-6) in the pathogenesis of collagen-induced arthritis Methods. CIA was induced by immunizing twice at a 3-week interval with bovine type I1 collagen (CII) emulsified with complete adjuvant. Rat anti-mouse IL-6 receptor (anti-IL-6R) monoclonal antibody MR16-1 or isotype-matched control antibody KH-5 was then injected once intraperitoneally. Symptoms of arthritis were evaluated with a visual scoring system, and serum anti-CII antibody and IL-6 levels were measur… Show more

“…Pathogenicity has been associated with the production of inflammatory cytokines, including TNF␣, IL-6, and IL-17 (29)(30)(31). Herein we identified IL-21 as a new T cell cytokine that is contributing to the pathologic processes of both CIA and AIA.…”

“…IL-21 has been reported to promote production of IL-17 from activated human T cells (41).Thus, blockade of IL-21 in CIA may function, in part, by modulation of IL-6-driven inflammation, either directly or through down-regulation of TNF␣ or IL-17. IL-6 has a pathogenic role in CIA (26,31,42) and is a critical inflammatory cytokine in RA. Blockade of IL-6R has demonstrated efficacy in clinical trials of RA (43).…”

“…The anti-IL-6 receptor tocilizumab has recently been approved for RA in Europe (39), and anti-IL-6 receptor treatment shows efficacy in CIA (40,41). The efficacy achieved in RA by blocking IL-6 receptor suggests that compounds targeting JAK-1 in addition to JAK-3 may have efficacy, at least in part, due to IL-6 inhibition.…”

“…The potency of IL-6 receptor blockade was subsequently tested in animal models of arthritis. One intraperitoneal injection of a monoclonal antibody recognizing the IL-6 receptor on day 0 or day 3 suppressed the development of murine CIA (106). Treatment with an antibody targeting IL-6 itself was also shown to reduce the incidence and severity of CIA in mice (107).…”
