Bleomycin-induced cutaneous toxicity in the rat: analysis of histopathology and ultrastructure compared with progressive systemic sclerosis (scleroderma

Abstract: Rats injected with bleomycin over a 58-week period developed weight loss, alopecia, hyperpigmentation, skin thickening and skin tautness when compared with saline-injected control animals. The only significant abnormality in laboratory blood tests was an increased sedimentation rate in the bleomycin-treated rats compared with controls. Histological examination of dorsal skin showed atrophied sebaceous glands and increased collagen fibres, with diameters ranging from 37.5 to 75 nm as compared with 72.5 to 100 n… Show more

“…Schleimhaut v.a. Hyperpigmentierungen, Hyperkeratosen, Exantheme, Alopezien, Ulzerationen oder Stromatitiden; selten infiltrierte Plaques oder Knoten bis hin zu sklerodermiformen Indurationen der Haut [1,2,5,11,16,19], wobei die mittlere hauttoxische Dosis in Studien mit vergleichsweise großen Patientenkollektiven mit etwa 165 mg angegeben wird [10,18].…”

“…One of the better-known models is bleomycin-induced scleroderma. A repeated intradermal or subcutaneous injection of bleomycin into rats and mice leads to the development of dermal sclerosis (Mountz et al, 1983;Yamamoto;Yamamoto & Nishioka, 2001Yamamoto, Takahashi, Takagawa, Katayama, & Nishioka, 1999). The latter is microscopically characterized by the deposition of thick collagen bundles, homogeneous acellular material, and cellular infiltrates of T-cells, macrophages, and mast cells.…”

“…Among other toxicities in humans, cutaneous toxicity including hyperpigmentation, alopecia, sclerosis, gangrene, edema, and fibrosis has been reported at higher incidence (14). BLM has been reported to be concentrated in the lung and skin due to the lower activity of the BLM-inactivating enzyme in these tissues (15,28 (Table II).…”
