Eco-toxicity of Neonicotinoid: a case study on the impact of imidacloprid using Drawida willsi earthworm as bioindicator

Many pesticides are used in the world and in India to boost the crop productivity, among which imidacloprid; a neonicotinoid is widely used now a day due to its low toxicity. These pesticides no doubt have protected the crops from the dangerous pest but in turn are becoming dangerous for soil health. Therefore an experiment was conducted to find out the eco-toxicity of imidacloprid. For this, earthworm (Drawida willsi, Michaelsen) and soil was collected from such agricultural field where there had no record of input of agrochemicals. Different concentrations of imidacloprid were prepared in dilution with acetone and sprayed on the soil surface. Five replicates for each concentration of the pesticides were prepared. Then ten numbers of juvenile, immature and adult earthworms were added separately into all the replicates of different concentrated samples. All the samples were kept in the laboratory under close vigil for 96 hours. Number of earthworm death with respect to doses and replicates were recorded and the 96h LC50 values for juvenile, immature and adult earthworm were calculated by Finney's Probit Method (Finney, 1971). It was found that the 96 h LC50 values with their 95% confidence limit of juvenile, immature and adult earthworm were 4.43, 7.96 and 12.45 mg a.i. imidacloprid/kg dry soil respectively. Although the recommended dose of imidacloprid was lower than the 96 h LC50 values of D. willsi for imidacloprid, but still it could affect the soil biota by altering its vital rates and metabolism due to bioaccumulation of the agrochemicals.

Glyphosate is a broad spectrum weed resistant herbicide. Glyphosate may pose negative impact on land ecosystems because of wide broad usage and hydrofilic characteristic. The aim of this study was to investigate negative effects of glyphosate on soil invertebrate organisms (earthworm Eisenia fetida). The duration of experiment was 8 weeks. The range of the test concentrations of glyphosate were: 0,1, 1, 5, 10, 20 mg/kg. To investigate the glyphosate impact on earthworm Eisenia fetida the following endpoints were measured: survival, reproduction and weight. The exposure to 20 mg/kg glyphosate has led to the 100% mortality of earthworms. Glyphosate has led to decreased E. fetida reproduction, the cocoons were observed only in the lowest concentration (0,1 mg/kg). In general: long-term glyphosate toxicity to earthworms (E. fetida) may be significant. Glifosatas yra herbicidas, plačiai naudojamas piktžolėms naikinti. Dėl masinio ir netinkamo naudojimo bei didelio tirpumo vandenyje glifosatas turi įtakos sausumos ekosistemoms. Eksperimento tikslas – įvertinti skirtingų glifosato koncentracijų poveikį žieduotosioms kirmėlėms (kompostiniam sliekui Eisenia fetida L.). Tyrimas buvo vykdomas 8 savaites. Eksperimento metu buvo naudotos 0,1, 1, 5, 10, 20 mg/kg glifosato koncentracijos dirvožemyje. Glifosato poveikio kompostiniams sliekams E. Fetida įvertinti buvo tirti augimo, mirtingumo ir reprodukcijos rodikliai. Eksperimento metu nustatyta, jog didelės glifosato koncentracijos letaliai veikė kompostinį slieką. Esant didžiausiai 20 mg/kg koncentracijai, sliekų mirtingumas jau pirmąją savaitę buvo 100 %. Didėjanti glifosato koncentracija mažino kompostinio slieko E. fetida svorio prieaugius bei neigiamai veikė sliekų reprodukciją, kokonai užfiksuoti tik esant mažiausiai koncentracijai. Eksperimento pabaigoje nustatyta tendencija – kuo ilgesnis glifosato ekspozicijos laikas, tuo didesnė žala daroma dirvožemio bestuburiams (Eisenia fetida).

Despite several known beneficial attributes, biochar is suspected to cause harm to soil organisms when present in relatively high quantities in the soil. To determine the potential detrimental effects of biochar, for 96 h, we exposed the earthworm Eisenia fetida to 0, 2, 4 and 8 mg glyphosate (GLY) per kg in non-amended and biochar-amended soil at rates of 5, 10 and 15%. The results indicated that in non-amended soil, survival was significantly decreased in the highest GLY concentration. Although no median lethal concentration (LC50) could be computed due to the lack of sufficient mortality, in the absence of biochar, a lethal concentration 10% (LC10) of 5.540 mg/kg and a lethal concentration 20% (LC20) of 7.067 mg/kg were calculated. In the biochar-amended soil, no mortality occurred in the control and GLY treatments for all three biochar amendment rates. Biomass results showed significant biomass loss in the highest GLY treatment in the absence of biochar, with an effective concentration of 10% (EC10) of 5.23 mg/kg and an effective concentration of 20% (EC20) of 6.848 mg/kg. In the amended soil, overall, slight non-significant increases in biomass were recorded and no effective concentrations could be calculated due to the lack of significant biomass loss. The assessment of neurotoxicity via the activity of acetylcholine esterase (AChE) showed no change in AchE due to GLY in all the non-amended treatments. However, in the biochar-amended treatments, statistically high levels of AchE occurred (p < 0.05) even in the control (in the absence of GLY). The assessment of oxidative stress through catalase (CAT) activity, showed similar results with no significant effects of GLY alone on CAT activity, but rather dramatic increases in activity in the control and GLY treatments in the biochar-amended soil, with one significant increase in the 10% amended in 8 mg GLY/Kg (p < 0.05). Such significant increases in both AChE and CAT were only observed in soil amended with 10 and 15% biochar. Our findings show that although seemingly beneficial for whole body endpoints, biomarker responses indicate that a biochar amendment higher than 5% adds considerable additional stress to earthworms and should be avoided.
