Biosynthesis of anthraquinones by interspecies cloning of actinorhodin biosynthesis genes in streptomycetes: clarification of actinorhodin gene functions

Abstract: Streptomyces galilaeus ATCC 31133 and ATCC 31671, producers of the anthracyclines aclacinomycin A and 2-hydroxyaklavinone, respectively, formed an anthraquinone, aloesaponarin II, when they were transformed with DNA from Streptomyces coelicolor containing four genetic loci, actI, actIll, actlV, and actVII, encoding early reactions in the actinorhodin biosynthesis pathway. Subcloning experiments indicated that a 2.8-kilobasepair XhoI fragment containing only the actl and actVII loci was necessary for aloesapona… Show more

“…Thus, from 20-L fermentation of Streptomyces sp. SB3501 in medium M1, we subsequently isolated six additional IFQs, named IFQ B-G (2-7), three IIQs, named IIQ A-C (8-10) and a known anthraquinone desoxyerythrolaccin (11), 18 together with 1 as the major metabolite.…”

“…Interestingly, in additional to be isolated from both plants and microorganisms, 26,27 11 has also been produced by a recombinant Streptomyces strain expressing the actI/actVII/actIV genes that encoded the type II polyketide synthase and associated cyclases for actinorhodin biosynthesis. 18 Since similar acyl carrier protein-tethered octaketide intermediates have been well established for actinorhodin biosynthesis, 18 11 could be viewed as a shunt product of 12 (Figure 2b), the isolation of which provided further support for the proposed pathway for 1-6 biosynthesis in Streptomyces sp. SB3501.…”

“…There is putative translational coupling between the following pairs of genes: actl-ORF1-actl-ORF2, actl-ORF3-actVII, and actVII-actIV. actVII mutants produce a shunt product called mutactin (31), whereas actVI mutants produce aloesaponarin II as a shunt product (1). Dehydromutactin, the dehydration product of mutactin, is also shown (see the text for details).…”
